摘 要:目的观察缺氧诱导因子1α(HIF-1α)和血管内皮生长因子(VEGF)在骨肉瘤组织中的表达及其与血管生成的关系。方法应用免疫组织化学(SP)法,检测35例骨肉瘤、14例骨软骨瘤中HIF-1α、VEGF的表达情况,同时用CD34标记血管内皮细胞,计算微血管密度(MVD)。结果 HIF-1α蛋白染色主要表达于细胞核,在骨肉瘤中阳性表达率高于在骨软骨瘤中阳性表达率(P<0.05)。VEGF蛋白染色主要表达于细胞质及细胞膜,在骨肉瘤中阳性表达率高于在骨软骨瘤中阳性表达率(P<0.05)。Ⅲ期骨肉瘤的HIF-1α、VEGF蛋白的表达强度与Ⅱa期、Ⅱb期比较差异有统计学意义(P<0.05),而Ⅱa期与Ⅱb期的表达强度比较差异无统计学意义(P>0.05)。35例骨肉瘤标本中有14例(40.0%)HIF-1α和VEGF的表达同时呈阳性,13例(37.1%)HIF-1α和VEGF的表达同时呈阴性,8例(22.9%)HIF-1α和VEGF的表达呈不一致性。HIF-1α和VEGF在骨肉瘤中的表达呈正相关。HIF-1α(+)、VEGF(+)的MVD值均显著高于HIF-1α(-)、VEGF(-)(P<0.01)。结论 HIF-1α和VEGF在骨肉瘤组织中均为高表达,提示它们可能在骨肉瘤新生血管生成中具有协同作用,对骨肉瘤的发生、发展起到重要作用。Objective To investigate the role of HIF-1α and VEGF in angiogenesis of osteosarcoma by analyzing their expression. Methods The expression of HIF- 1α and VEGF specimens of 35 osteosaroma and 14 osteochondroma tissues was evaluated by immunohistochemistry. MVD was calculated by using CD34 as the specific marker of microvessel endothelium. Results HIF- 1α protein staining was mainly expressed in the nucleus. The positive rate of HIF- 1а in osteosarcoma was significantly higher than those in osteochondroma tissues(P < 0. 05).VEGF protein staining was mainly expressed in the cytoplasm and cell membrane. The positive rate of VEGF in osteosarcoma was significantly higher than those in osteochondroma tissues(P < 0. 05). There was significant difference between phase Ⅲ and phase Ⅱa and Ⅱb on the expression intensity of HIF- 1α and VEGF in osteosarcoma(P <0. 05). There was no difference between phase Ⅱa and phaseⅡb on the expression intensity of HIF- 1α and VEGF in osteosarcoma(P > 0. 05). The positive simultaneously rate of HIF- 1α and VEGF was 40. 0% in 35 cases of osteosaroma. The negative simultaneously rate of HIF- 1α and VEGF was 37. 1% in 35 cases of osteosaroma. 8 cases(22. 9%) HIF-1α and VEGF expression were inconsistencies. The expressions of HIF-1α and VEGF in osteosarcoma were positively correlated(P < 0. 05). The expression of HIF- 1α and VEGF were correlated with staging(P <0. 05). The MVD was higher in the positive group of HIF- 1α and VEGF than their negative group(P < 0. 05). Conclusion s The positive expression of HIF- 1α and VEGF are related with osteosarcoma microvascular generate. It seems that HIF- 1α and VEGF contribute to development of osteosarcoma.
