摘 要:研究采用微波辅助法提取灵杆菌脂多糖的工艺。在比较灵杆菌脂多糖的传统提取方法与微波辅助提取法的基础上,采用单因素试验分别考察微波处理时间、微波处理功率、固液比、浸提时间对灵杆菌脂多糖提取效果的影响,并通过L9(34)正交试验,优化微波辅助提取BP-LPS的工艺参数。结果表明,微波辅助提取灵杆菌脂多糖的最佳工艺条件为:提取固液比1∶30,微波处理3 min,功率250 W,浸提60 min。在此条件下,灵杆菌脂多糖提取量最高达13.413 mg/g,相比传统提取方法提高99.4%,证明微波辅助提取是一种简便、高效的提取方法。To study the process of microwave-assisted extraction for bacterium prodigiosum lipopolysaccharides, this study used single factor test and investigated the influence of four factors on polysaccharides extraction, including microwave processing time, microwave processing power, material/liquid ratio and extraction time, which were based on the comparison between traditional extracting methods and microwave-assisted method. And then through L9 ( 3^4 ) orthogonal test, the optimum parameters of microwave assisted extraction BP - LPS were determined. The test results indicated that the best extraction conditions were as follows : the micro- wave treatment time of 3 min, microwave power of 250 W, the material/liquid ratio of 1 : 30 and the extraction time of 60 min. Under these conditions, the total yield of polysaccharide was 13.413 mg/g, which was increased by 99.4% compared with the traditional extraction method. Therefore,it is found that microwave-assisted extraction is a simple and efficient extracting method.
