内皮素拮抗剂抑制人源性喉癌Hep-2细胞VEGF-C体外表达的定量分析

摘　　要：通过影响人源性喉癌Hep-2细胞中血管内皮生长因子C（VEGF—C）表达，探讨内皮素拮抗剂（BQ-123）与喉癌淋巴转移的机制，为临床及基础研究提供相应的实验依据．体外培养人源性喉癌Hep-2细胞，运用酶联免疫吸附法（ELISA）测定细胞培养液中VEGF—C的含量；利用M1-r比色法，测定不同浓度的内皮素拮抗剂对人源性喉癌Hep-2细胞增殖的影响．实验中发现当浓度分别为20．0～150．0μg／mLBQ-123作用48h，对人源性喉癌Hep-2细胞增殖有显著抑制作用（P〈0．05）；浓度分别为50．O～150．0μg／mLBQ-123对人源性喉癌Hep-2细胞VEGF—c蛋白分泌量有明显抑制活性作用（P〈0．05）．BQ-123可以有效抑制人源性喉癌Hep-2细胞中VEGF—C体内的表达．Through the study of endothelin receptor blockade about the vascular endothelial growth factor C （VEGF-C） in the human laryngeal carcinoma Hep-2 cells, the mechanism between BQ- 123 and the lym-phatic metastasis is discussed. In vitro Hep - 2 cells are cultivated. The content of VEGF-C in the culture fluid is determined by ELISA. MTT is then adopted to examine the influence of endothelin receptor blockade with dif-ferent doses on the proliferation of human laryngeal carcinoma Hep-2 cells. It is found that it has significantly higher inhibition on Hep -2 ceils when it is 20.0 - 150. 0 μg/mL with BQ - 123 after 48h（P 〈0.05） ; and that the activation of VEGF - C secretion in Hep - 2 cells is reduced remarkably with 50.0 - 150.0μg/mL BQ - 123. It is pointed out that BQ - 123 can significantly inhibit the expression of VEGF - C in Hep-2 cells, which may become a potential treatment for human laryngeal carcinoma.
