摘  要:目的:研究MHV-3(鼠Ⅲ型肝炎病毒)感染所致的暴发性肝炎小鼠模型中,IL(白细胞介素)-9在肝脏NK(自然杀伤细胞)及NKT细胞的表达变化,及其与肝损伤之间的关系。方法:BALB/cJ小鼠腹腔注射100pfu(空斑形成单位)MHV-3建立小鼠暴发性肝炎模型,应用流式细胞术荧光分析法检测MHV-3感染0小时、24小时、48小时和72小时肝脏NK及NKT细胞表达IL-9的比例变化,并检测小鼠血清中丙氨酸转氨酶(ALT)水平。结果:BALB/cJ小鼠感染MHV-3后,IL-9在肝脏NK细胞的表达在感染24小时显著升高(P<0.05),并于48小时达最高水平(P<0.05);在肝脏NKT细胞的表达于感染24小时显著升高(P<0.05),并于48小时达最高水平,且均与肝损伤指标(血清ALT水平)呈正相关。结论:随着感染时间延长,肝脏分泌IL-9的NK和NKT淋巴细胞显著升高,提示IL-9参与了MHV-3诱导的暴发性肝炎小鼠免疫诱导的肝脏损伤发病过程。Objective: To investigate the changes of hepatic IL-9-secreting NK and NKT cells in MHV-3-induced fulminant hepatitis in mice and their correlation with liver injury. Methods: MHV-3-induced fulminant hepatitis model was established in mice. The percentage of hepatic IL-9-secreting NK and NKT cells were determined by flow cytometry, and the level of ALT in se- rum were examined. Results: By the time-colirse of MHV-3 infection, the percentage of hepatic IL-9-producing NK cells in- creased at 24h ( P 〈 0.05 ) and reached peak at 48h with great significance, while the percentage of hepatic IL-9-producing NKT cells increased at 24h (P 〈 0.05 ) and reached peak at 48h with no significance. The percentage of hepatic IL-9-producing NK and NKT cells were both positive correlated to the index of liver injury (serum ALT) . Conclusion: The increased expression of IL-9 from hepatic NK and NKT lymphocytes may contribute to immune-mediated liver injury in MHV-3-induced fulminant hepati- tis in mice.
