摘  要:目的探讨WNT10A在肾细胞癌(RCC)中的表达及作用机制。方法收集1998年1月-2006年12月诊治的RCC患者284例和良性肾疾病(BRD)患者267例的完整病历资料,各组随机抽取病理样本,RT-PCR法筛选差异表达WNT配体,免疫组化法检测WNT/β-catenin的信号通路蛋白。结果 RT-PCR结果表明,WNT家族中WNT1、WNT2、WNT3A、WNT8A、WNT9A、WNT10A在RCC组中呈高表达,分别高于BRD组2.51、1.85、1.98、2.22、2.30和9.12倍,差异有统计学意义(P<0.05)。免疫组化染色结果显示,RCC组WNT10A、β-catenin、c-myc、CyclinD1蛋白表达明显高于BRD组,差异有统计学意义(P<0.05,P<0.01)。结论 WNT10A癌基因通过激活WNT/β-catenin的信号通路在RCC的发生和进展中发挥重要作用。Objective To determine the role of WNT10A gene in renal cell carcinoma(RCC) and underlying mechanisms. Methods The study accrued subjects in a 9 year follow up cohort comprising RCC and benign renal disease(BRD) patients from Jan 1998 to Dec 2006,and the complete medical records were also collected.After the pathological samples were chosen randomly from selected cases,the expression of WNT family ligands were screened using RT-PCR,and the proteins of WNT/p-catenin signaling pathway were detected by immunohistochemical method.Results PCR results showed that the expression of WNT1,WNT2,WNT3A,WNT8A,WNT9A,WNT10A gene in RCC tumor tissue was 2.51,1.85,1.98,2.22,2.30 and 9.12 times higher than that in normal renal tissue(P 0.05).Immunohistochemical results showed that compared with normal kidney tissue,the expressions of WNT10A,p-catenin,c-myc and Cyclin Dl in RCC tumor tissue were increased sig- nificantly(P 0.05,P 0.01).Conclusion This study suggests that WNT10A acts as an oncogene both in RCC carcinogenesis and progression by activating WNT/β-catenin signaling.
