摘　　要：目的:探讨Notch信号分子及下游靶基因在人NK/T细胞淋巴瘤、NK细胞淋巴瘤、T细胞淋巴瘤和B细胞淋巴瘤细胞中的表达,阐明Notch信号分子在NK细胞肿瘤中的作用及意义。方法:选择NK/T细胞淋巴瘤细胞SNK-6、NK细胞淋巴瘤细胞YTS、人急性T淋巴细胞白血病细胞Jurkat和人Burkitt’s淋巴瘤细胞Raji,通过实时荧光定量PCR技术检测上述细胞中Notch信号分子的表达水平。结果:Notch 1,3,4信号分子在YTS和SNK-6细胞中表达量均不同程度高于Raji和Jurkat淋巴瘤细胞(P<0.01),但是在SNK-6中的表达量均不同程度低于在YTS细胞中的表达(P<0.01)。Notch 2信号分子在YTS、SNK-6细胞中的表达量高于Jurkat细胞(P<0.01),但是与Raji细胞比较差异无统计学意义(P>0.05)。Notch下游靶基因Hes1的表达量,YTS细胞高于Raji和Jurkat细胞(P<0.05,P<0.01),而SNK-6细胞则低于Raji和Jurkat细胞(P<0.01)。结论:在NK细胞肿瘤细胞中Notch信号分子表达异常,且明显区别于B、T细胞淋巴瘤细胞。Objective To investigate the expressions of Notch signal molecules and their downstream target genes in human NK/, NK,T and B lymphoma cells and to expound the effect and significance of Notch signal molecules in NK cell tumor.Methods The expression levels of Notch signal in YTS,SNK-6,Raji and Jurkat cells were detected by Real-time PCR.Results The expression levels of Notch 1,3,4signal molecules in YTS and SNK-6 cells were significantly higher than those in Raji and Jurkat cells （P0.01）, but the expression levels of Notch 1,3,4signal molecules in SNK-6cells were lower than that in YTS cells （P0.01） .The expression levels of Notch 2in YTS and SNK-6cells were significantly higher than that in Jurkat cells （P0.01）, but there was no statistically significant difference compared with Raji cells （P0.05） .The expression level of Hes1in YTS cells was significantly higher than those in Raji and Jurkat cells （P0.05or P0.01）, but the expression level in SNK-6cells was lower than those in Raji and Jurkat cells （P0.01） .Conclusion Notch signal molecules express abnormally in NK cell tumor,and they are significantly different with B and T lymphoma cells.
