摘  要:目的:鉴定一种新的检测乳腺癌循环癌细胞的肿瘤标记物FAM83A。探讨FAM83A在乳腺癌外周血中的表达水平及其与临床病理学特征的关系。方法:通过肿瘤基因组解剖计划(cancergenome anatomy project,CGAP)数据库的数字基因表达演示工具(digital gene expression displayer,DGED),发现了一种新的乳腺癌外周血标记物FAM83A。采用实时半定量巢式PCR技术,检测了84例乳腺癌和60例正常人外周血中FAM83A的表达水平,并进一步分析了FAM83A表达与乳腺癌临床病理特征的相关性。结果:发现了一个新的外周血肿瘤标记物FAM83A,并证实其在84例乳腺癌外周血中的表达水平明显高于正常组(P<0.01)。并且,FAM83A在外周血中的表达水平与临床分期、淋巴结转移和ER、PR具有相关性(P<0.05)。结论:FAM83A是一种新的检测乳腺癌外周血循环癌细胞的肿瘤标记物,并可作为判断乳腺癌转移和预后的评估指标。Objective: To identify a new tumor marker FAM83A for detection of circulating cancer cell in breast cancer patients.To investigate the relationship between the expression of FAM83A and the clinicalpathological features in breast patients.Method: A new tumor marker FAM83A for detection of circulating cancer cells was identified by the DGED tool in the CGAP.We detected the expression level of FAM83A in 84 breast cancer patients and 60 healthy volunteers by real-time semiquantitative nested PCR and then analysed the correlation of expression of FAM83A and clinicalpathological features.Result: A new tumor marker FAM83A for detection of circulating cancer were found,and the marker was highly expressed in cancer patients than normal control ones(P0.01).High-level expression of FAM83A positively correlated with clinical stages,lymph nodal metastasis and ER,PR(P0.05).Conclusion: FAM83A,a new tumor marker for detection of circulating cancer cells,may serve as useful markers to predict metastasis and to evaluate prognosis of patients with breast cancer.
