外周血MAGE-1基因表达与非小细胞肺癌复发转移关系的研究

摘  要:目的探讨外周血黑色素瘤抗原-1(MAGE-1)基因表达与非小细胞肺癌(NSCLC)转移、复发的关系。方法 56例NSCLC患者,34例肺部良性疾病患者(对照组),采用逆转录-聚合酶链反应(RT-PCR)检测血液中MAGE-1 mRNA表达水平,并对NSCLC患者进行随访。结果 NSCLC组患者外周血中的MAGE-1mRNA阳性表达率分别为56.86%(29/56),对照组34例均无表达。术后2年,51例无远处转移患者出现复发、转移26例,其中I期1例,Ⅱ期4例,ⅢA期11例,ⅢB期10例。外周血MAGE-1 mRNA表达阳性组与MAGE-1 mRNA表达阴性组的复发转移比较差异无统计学意义(P>0.05)。结论 MAGE-1可作为非小细胞肺癌微转移的肿瘤标记物,外周血MAGE-1 mRNA表达阳性与NSCLC肿瘤复发(转移)无关联。Objective To explore the relationship of expression of melanoma antigen 1(MAGE-1) gene in the peripheral blood with reoccurrence and metastasis of non-small cell lung cancer(NSCLC).Methods Fifty-six NSCLC patients and 34 patients with benign pulmonary diseases(control group) were enrolled in the study.Reverse transcription-polymerase chain reaction(RT-PCR) was used to detect the expression level of MAGE-1 mRNA in blood.A follow-up was conducted in the NSCLC patients.Results The positive expression rate of MAGE-1 mRNA was 56.86%(29 /56) in the peripheral blood of NSCLS patients,no positive expression of MAGE-1 mRNA was observed in the control group.Two years after operation,there were 26 cases of tumor reoccurrence and metastasis in 51 cases of non-distal metastasis,including 1 case of Ⅰstage,4 cases of Ⅱ stage,11 cases of ⅢA stage,10 cases of ⅢB stage.The expression of MAGE-1 mRNA had no relation with tumor recurrence or metastasis(P 0.05).Conclusion MAGE-1 can be used as a molecular marker for the detection of NSCLC micrometastasis.The positive expression of MAGE-1 mRNA in the peripheral blood of NSCLC has no relation with tumor recurrence or metastasis.
