摘 要:目的观察安神定志灵对注意缺陷多动障碍(ADHD)动物模型-自发性高血压大鼠(SHR)纹状体多巴胺转运体(DAT)表达的影响,探讨该药治疗ADHD的作用机制。方法 SHR大鼠随机分为模型对照组、安神定志灵高、中、低剂量组,Wistar-Kyoto(WKY)大鼠为正常对照组。安神定志灵高、中、低剂量组分别按生药剂量3.41、1.71、0.85g/mL灌胃;模型对照组和正常对照组以2mL/100g生理盐水灌胃,连续给药2周后处死大鼠,取脑用ELISA检测各组大鼠脑组织中儿茶酚-氧位甲基转移酶(COMT)含量。结果模型对照组、安神定志灵低、中、高剂量组与对照组比较具有显著性差异(P<0.01);安神定志灵低、中、高剂量组脑组织中COMT含量较模型对照组均降低,差异有显著性(P<0.01);安神定志灵中剂量组与高剂量组两组间COMT含量没有显著性差异(P>0.05);安神定志灵低剂量组与对照组两组间COMT含量差异没有显著性(P>0.05)。结论:SHR大鼠(ADHD模型鼠)脑组织中COMT含量存在改变,说明COMT在ADHD的发病中具有一定意义;安神定志灵对其有着调节作用。Objective To study the effects of Anshendingzhiling(ADL) an expression of dopamine transporter(DAT) of spontaneously hypertensive (SHR) rats' s striatum and the mechanism of this medicine to treat with ADHD. Method Thirty SHR rats were randomly di- vided into 5 groups including model group, ritalin group, ADL high dose group, ADL medium dose group and ADL low dose group,6 rats each group, meantime ,6 Wistar - Kyot rats constitute normal control group. The dosages of the 3 groups of ADL were 34.1,17.1,8.5 g kg - 1, ig respectively and the dosage of ritalin group is 2.1 mg kg - 1, ig, as for the model group or normal control, rats were given physiological saline as per 10 mL kg - 1 ,ig. After 2 - week,the rats in experiments were killed and separate the brain tissues, then examine the COMT of each group with ELISA. Result Compared with normal group, the significant differences in model group,high dose of ADL group, middle dose of ADL group and low dose of ADL group( P 〈 0.01 ). The content of COMT of brain tissue is significant lower in all levels of ADL(P 〈 0.01 ). The content of COMT is no difference between high dose and middle dose of ADL group( P 〉 0.05). The content of COMT is no difference be- tween low dose of ADL group and the normal group(P 〉 0.05). Conclusion The change of COMT content of SHR rats' brain tissue show the COMT plays an important role in ADHD incidence, and the ADL has a moderating effect of it.
