摘  要:目的探讨尼克酰胺腺嘌呤二核苷酸磷酸(NADPH-d)在大鼠三叉神经节(TG)和三叉神经中脑核(MTN)中的表达。方法大鼠常规麻醉、固定后,参照Paxinos和Watson的大鼠脑立体定位图谱,用冷冻切片机于Bregma-9.8mm和Bregma-7.4mm之间作MTN冠状切片,TG作水平切片,片厚均为20μm。每隔4张取一张,采用NADPH-d组织化学染色及计算机图像分析系统对所取脑片进行观察。结果在生理条件下,TG的初级感觉神经元大部分呈现阳性反应,在TG的神经元之间可见NADPH-d阳性神经纤维,形成细胞周围的篮样结构;NADPH-d阳性神经元在整个MTN从尾端到头端都有表达,有许多NADPH-d阳性神经纤维组成了稠密的丛。结论 NADPH-d在TG和MTN中高度表达,表明大鼠TG和MTN的三叉神经的传入信息是由硝基能神经纤维传入。推测NO可能在口腔颌面部的疼痛和本体感觉中起到一定的作用。Objective To study the expressions of NADPH-d of the trigeminal primary afferent neurons located in the trigeminal ganglion (TG) and mesencephalic trigeminal nucleus (MTN) in rats. Methods The rats were anesthetized and im- mobilized as routine. Referring to Paxinos and Watson rat stereotaxic atlas, a frozen section was made with the thickness of 20 μm from Bregma--9.8 mm to Bregma--7.4 mm. The brain slices were observed with a light microscope after NADPH-d histochemieal stain and computer image analysis. Results In TG, most of the neurons were NADPH-d labled neurons, and NADPH-d-posi- rive nerve fibers dispersed throughout the ganglion forming basket-like around the neurons; in MTN, from the caudal to the cranial part, the positived-neurons were expressed. There were a lot of NADPH-d-positive nerve fibers, which formed dense plexus. Conclusion Strong expression of NADPH-d in TG and MTN suggest that trigeminal sensory information processing in the rat is controlled by nitrergie input. NO may play a role in the pain transmission and proprioceptive information from the orofacial in the rat trigeminal afferent pathways.
