摘 要:目的探讨肿瘤坏死因子-α(TNFα)对人宫颈癌细胞HeLa侵袭能力的影响及其分子机制。方法 5ng/mL TNFα处理HeLa细胞,体外侵袭转移实验观察TNFα诱导后细胞的侵袭能力;免疫荧光及蛋白印迹观察核转录因子-κB p65(NF-κB p65)的定位及表达,蛋白印迹法检测基质金属蛋白酶9(MMP9)的表达;使用NF-κB通路抑制剂Bay11-7082(2.5μmol/L)处理24h后TNFα(5ng/mL)诱导细胞,观察细胞侵袭能力及MMP9表达的改变。结果 TNFα促进了HeLa细胞的侵袭,诱导NF-κBp65核转位激活,上调了MMP9的表达。TNFα促进HeLa细胞的侵袭能力及MMP9的表达可以被NF-κB抑制剂阻断。结论 TNFα通过激活NF-κB信号促进了宫颈癌细胞侵袭,暗示NF-κB可能作为抑制宫颈癌进展的重要靶点。Objective To explore the effects and molecular mechanisms of tumor necrosis factor-α (TNF-α) on the invasion of human cervical cancer Hela cells. Methods Hela cells were cultured in medium containing TNFα (5 ng/mL) and their invasiveness was investigated in vitro by Millicell assay; NF-κB activity of Hela cells was detect by immunofluorescent assay and Western blotting. MMP9 expression was examined by Western blot. Furthermore, NF-kB activation inhibitor Bay11-7082 was used to confirm the role of NF-kB activation induced by TNFα. Hela cells pretreated with or without Bay 11-7082 (2.5 μmol/L) for 24 h and then treated with or without TNFα (5 ng/ mL) were observed for invasion and MMP expression. Results TNFα promoted the invasive ability of Hela cells, induced the activation of NF-κB, and up-regulated MMP9 expression. Moreover, NF-κB inhibitor blocked TNFα- enhanced invasion and MMP9 expression. Conclusion Activation of NF-κB induced by TNFα promotes the invasion of cervical cancer cells, suggesting that NF-κB may be a potential target for suppressing the metastasis and invasion of cervical carcinoma.
