摘  要:目的探讨抗增殖蛋白(PHB)在宫颈鳞状细胞癌(SCC)中的表达及意义。方法用免疫组化SP二步法检测了57例宫颈组织标本中PHB的表达,其中SCC 20例、宫颈上皮内瘤样病变(CIN)17例、慢性宫颈炎20例。采用酶联免疫吸附试验(ELISA)测定50例血清标本中PHB的表达,其中SCC 14例,CIN 3级10例,正常人26例。结果免疫组化结果显示,PHB在SCC、CIN及慢性宫颈炎组织中的阳性率分别为75%、52.9%和15%,3组间的差异有统计学意义(P<0.05),且其阳性表达与宫颈病变程度呈正相关(rs=0.949,P<0.05);PHB在SCC、CIN患者和正常人血清中的质量浓度中位数分别为:31.41、28.53、76.94ng/mL,PHB在正常人血清中的表达显著高于其在CIN 3级及SCC中的表达(P<0.05)。结论 PHB在不同宫颈病变中存在差异表达,有可能成为宫颈癌的分子标志物。Objective To investigate the expression and significance of prohibitin(PHB)in cervical squamous cell carcinoma (SCC). Methods PHB expression was detected in 57 cervical tissue samples from 20 SCC, 17 cervical intraepithelial neoplasia (CIN) and 20 chronic cervicitis cases by immunohistochemical streptavidin-peroxidase (SP) method. Meanwhile, serum concentrations of PHB were measured by enzyme-linked immunosorbent assay (ELISA) in 14 patients with SCC, 10 patients with CIN 3 and 26 healthy women. Results Immunohistochemical staining showed that positive expressions of PHB were 75%, 52.9% and 15% in tissues from cervical cancer, CIN and chronic cervicitis, respectively (H=14.483, P〈0.05). The expressions were positively correlated with the grade of cervical lesions (rs=0.949, P〈0.05). ELISA showed that the medians of serum PHB concentrations were 31.41ng/mL, 28.53ng/mL and 76.94ng/mL, respectively, in patients with SCC and CIN and healthy women. The serum concentrations were significantly lower in cervical cancer and CIN patients than those in the healthy women (H=13.085, P〈0.05). Conclusion PHB is expressed differently in cervical lesions and may be a potential biomarker of SCC.
