10×Aβ_(3~10)基因重组腺病毒疫苗免疫BALB/c鼠的脾细胞增殖反应

摘  要:目的研究10×Aβ3~10基因重组腺病毒疫苗在BALB/c鼠体内产生的炎症免疫反应。方法 14只3月龄雄性小鼠按随机数字表法分为目的基因组、空载体组、Aβ42组,接种相应疫苗。脾细胞培养,MTT法测各组脾细胞增殖率,ELISA法检测培养上清中细胞因子白细胞介素(IL)-4和干扰素(IFN)-γ水平。结果目的基因组和Aβ42组均在各自相应抗原重新刺激时表现出较高的脾细胞增殖率,即目的基因组在Aβ3~10多肽重刺激时脾细胞增殖率较高,而Aβ42组则在Aβ42重刺激时脾细胞增殖率较高;Aβ42组的脾细胞培养上清中IL-4和IFN-γ水平均高于空载体组,目的基因组培养上清中IL-4水平与Aβ42组相比差异无统计学意义,但IFN-γ水平显著低于Aβ42组(P<0.05)。结论 10×Aβ3~10基因重组腺病毒疫苗能够有效诱导Th2型免疫反应,而不引起针对Aβ42的T细胞免疫反应。Objective To investigate the immune response induced by adenovirus vaccine containing 10×Aβ3~10 in BALB/c mice.Methods Mice were randomly divided into groups vaccinated with gene-recombinated adenovirus containing 10×Aβ3~10,empty gene-recombinated adenovirus and Aβ42 peptide respectively.MTT was used to detect splenocytes proliferative responses,ELISA was used to analyze IL-4 and IFN-γin restimulated splenocytes supernate.Results A robust proliferative response was only observed in splenocytes restimulated with the corresponding immunogens,restimulation of splenocytes in 10×Aβ3~10 group with 10×Aβ3~10 peptide resulted in robust proliferative response while in Aβ42 group with Aβ42 peptide.Aβ42 group showed highest levels of IL-4 and IFN-γ of all three groups whereas 10×Aβ3~10 group showed equal level of IL-4 but significantly lower level of IFN-γ compared to Aβ42 group.Conclusions Gene-recombinated adenovirus vaccine containing 10×Aβ3~10 can induce significant Th-2 polarized immune response while avoiding T cell immune response directed at Aβ42.
