摘 要:目的探讨丙戊酸钠(VPA)对胃癌SGC-7901细胞的放疗增敏作用及其可能机制。方法 MTT法检测丙戊酸钠对胃癌SGC-7901细胞的抑制率,计算半数抑制浓度(IC50)。流式细胞仪(FCM)分析细胞周期及细胞凋亡率。结果 VPA作用后胃癌细胞SGC-7901的增殖受到抑制,凋亡增强,S期细胞比例减小,G1期细胞增比例增大,联合放疗组效果较单纯组更为显著;MTT实验结果显示VPA作用SGC-7901细胞24、48、72 h后IC50分别为19.83、3.67、2.75mmol/L。结论丙戊酸钠联合放疗可诱导SGC-7901细胞细胞周期阻滞和细胞凋亡,从而抑制SGC-7901细胞生长。Objective To investigate radiosensitization effect of valproic acid on gastric carcinoma cell line SGC-7901 and its mechanism.Methods Huma gastric carcinoma cell line SGC-7901 was tested by MTT assay for cell proliferation and concentration of IC50,Flow cytometry was adopted to analyze the changes in cell cycle and apoptosis.Results After treated with valproic acid,the cell proliferation was inhibited,cell apoptosis were increased,the proportion of S phase was reduced and the proportion of G1 phase were increased.The combination group had more significant effect on human gastric carcinoma cell SGC-7901 than the alone group.The IC50 of VPA treated SGC-790 cells was19.83 mmol/L in 24 h,3.67 mmol/L in 48 h and 2.75 mmol/L in 72 h.Conclusion The combination of VPA and radiotherapy could inhibit the growth SGC-7901 lines by means of inducing cell cycle arrest and apoptosis.
