摘 要:蛹虫草Cordyceps militaris是当前用于功能食品和药物开发上较受国内外关注的真菌之一。本研究通过1次一因素法和正交实验设计优化了蛹虫草产虫草菌素和菌丝体的深层培养工艺。最适合菌丝生长的pH值和温度为6.0和20℃,而最适合虫草素积累的pH值和温度为5.0和26℃。蔗糖、蛋白胨、MgSO4、K2HPO4和NAA分别是最适合虫草素积累的碳源、氮源、无机盐和生长因子;培养基的成分对蛹虫草生物量的影响次序为:蔗糖>蛋白胨>MgSO4·7H2O>K2HPO4>NAA;培养基的成分对蛹虫草产虫草素的影响次序为:蛋白胨>K2HPO4>NAA>MgSO4·7H2O>蔗糖。最适合蛹虫草菌丝生长的培养基配比为4%蔗糖,2.5%蛋白胨,0.05%MgSO4·7H2O,0.15%K2HPO4,2.0mg/l NAA;最适合蛹虫草深层发酵产虫草素的培养基配比为4%蔗糖,1.5%蛋白胨,0.05%MgSO4·7H2O,0.05%K2HPO4,4.0mg/l NAA的培养基;在最佳蛹虫草深层发酵产虫草素的工艺条件下,CM8菌株胞外虫草菌素产量达到961.21 mg/l,胞内虫草菌素的含量达到12.87mg/g;菌丝体的产量为22.97 g/l,本研究得出的结论对深层培养提高蛹虫草虫草菌素和菌丝体的产量有借鉴意义。This study was aimed to optimize the conditions of submerged culture for mycelial growth and cordyce- pin production of C. militaris by one-factor-at-a-time and orthogonal layout methods. The optimal initial pH value and temperature for mycelial growth were 6.0 and 20 ℃, while those for cordycepin production were 5.0 and 26 ℃, respectively. Among the medium components, sucrose, peptone, MgSO4, K2HPO4, NAA were the most suit- able carbon, nitrogen, mineral sources and growth factor. The effects of media composition on the mycelial growth of C. militaris were orderly sucrose 〉 peptone 〉 MgSO4 7H2O 〉 K2HPO4 〉 NAA, and those on cordycepin produc- tion were orderly peptone 〉 K2HPO4 〉 NAA 〉 MgSO4 7H20 〉 sucrose. The optimal concentration of the compo- nents for mycelial growth were 40 g/1 sucrose, 25 g/1 peptone, 0.5 g/1 MgSO4 7H2O, 1.5 g/1 K2HPO4, 2.0 mg/1 NAA, and those for cordycepin production were 40 a/1 sucrose. 15 g/Inentnna 0. 5 g/1 MgSO4 7H2O, 0.5 g/1 K2HP04, 4.0 rag/1 NAA. This optimization strategy in shake flask culture resulted in a mycelial yield of 22.97 g/l, and cordycepin pro- duction in culture broth and in cells were 961.21 mg/1 and 12.87 mg/g respectively. The results obtained in this work will have a potential impact on the industrial scale production of cordycepin by submerged culture.
