摘  要:目的:探讨中西药联合(班贞1号+5-FU+TMP)对胃癌SGC-7901/ADR细胞MRP、LRP基因表达的影响。方法:以胃癌多药耐药细胞株SGC-7901/ADR为靶细胞,以不加药组为阴性对照组,5-氟尿嘧啶(5-FU)组、班贞1号组、班贞1号+5-FU组、中西药联合组为实验组,采用半定量RT-PCR法检测SGC-7901/ADR细胞在不同药物作用下MRP、LRP mRNA的表达。结果:5-FU组与阴性对照组相比,MRP、LRP mRNA表达差异无统计学意义(P>0.05)。中西药联合组MRP、LRP mRNA表达明显降低,与阴性对照组及其他实验组比较有统计学意义(P<0.05)。结论:中西药联合能够抑制SGC-7901/ADR细胞MRP、LRP mRNA表达,逆转了SGC-7901/ADR细胞的耐药性,其机制可能与下调MRP、LRP基因表达有关。Objective : To explore the effect of combination of Tetra methylpyrazine ( TMP), Banzhen No. 1 and 5 - Fu on the expression of gene LRP and MRP of cells Line SGC -7901/ADR in human gastric cancer. Methods: SGC -7901/ADR cells were taken as target cells, non - medicine group was taken as the negative control group, 5 - FU group, Banzhen No. 1 group, Banzhen No. 1 and 5 - Fu group and the group of combination of modern medicine and traditional chinese medicine were taken as the experimental groups. Semi- quantitative Reverse Transcriptase Polymerase Chain Reaction (RT- PCR) was used to deter- mine the expression of gene LRP, MRP mRNA in the Cell Line SGC -7901/ADR with different medicines. Results: There was no significant difference in the expression of LRP and MRP mRNA between the 5 - FU group and the negative control group (P 〉 0. 05) . Compared with the negative control group and other experimental groups, the expression of gene LRP and MRP mRNA significantly decreased in the group of combination of modern medicine and traditional chinese medicine ( P 〈 0.05 ). Conclusion: Combination of modern medicine and traditional Chinese medicine can inhibit the expression of LRP and MRP mR- NA in Cell Line SGC -7901/ADR and reverse drug resistance of Cell Line SGC -7901/ADR, whose mechanism is likely to be correlated to decrease in the expression of LRP and MRP mRNA.
