摘  要:目的评价体外培养与鉴定行全飞秒激光小切口微透镜摘除术(small incision lenticule extraction,SMILE)的近视患者角膜基质层来源的人角膜成纤维细胞的可行性。方法 8例16眼行SMILE的近视患者的角膜基质层组织分别进行组织块培养,采用细胞形态学观察及免疫细胞化学法进行鉴定。结果 8例SMILE来源的基质层人角膜成纤维细胞全部原代培养成功并稳定传代,细胞呈长梭形或三角形;免疫细胞化学鉴定:波形蛋白染色阳性,而角蛋白、结蛋白、S-100染色均为阴性。结论 SMILE来源的近视患者的角膜基质层是人角膜成纤维细胞培养的方便的组织来源。Objective To evaluate the feasibility of culturing and identifying fi- broblasts derived from human corneal stroma of myopic patients that had underwent the femtosecond laser small incision lenticule extraction(SMILE) in vitro. Methods Cor- neal stromal tissues from 8 patients( 16 eyes) with myopia that had underwent femto- second laser SMILE were primarily cultured using explant tissue culture technique, re- spectively, and were then identified by morphological observation and immunocyto- chemical staining. Results All the primary culture of fibroblasts derived from human corneal stroma were successfully carried out and the cell passages were stable. The cells resembled long spindle or triangle. Immunocytochemical staining results showed that vi- mentin staining was positive, while keratin staining, desmin staining and S-100 staining were all negative. Conclusion Corneal stroma of myopic patients that had underwent femtosecond laser SMILE is a convenient origin of tissue for the culture of fibroblasts derived from human corneal stroma.
