摘  要:目的探讨放射对小肠黏膜上皮细胞IL-2和IL-6分泌的影响以及小牛血去蛋白提取物(商品名:爱维治)在抑制小肠上皮黏膜细胞凋亡中的作用。方法将50只大鼠随机分为正常组、放射组、低剂量爱维治组、中剂量爱维治组以及高剂量爱维治组,每组各10只。采用高能χ摄线直线加速器对放射组及低、中、高剂量爱维治组大鼠腹部进行照射,照射剂量为9.0 Gy,并建立大鼠辐射损伤模型。连续4 d对建模大鼠腹部注射用药,并取大鼠小肠黏膜上皮制成病理切片,应用全自动化分析仪检测各组大鼠小肠黏膜上皮细胞中IL-2以及IL-6水平,采用免疫组化法分析爱维治对小肠黏膜上皮细胞凋亡的影响。结果放射组大鼠小肠黏膜上皮细胞中IL-2、IL-6水平显著高于其他各组,差异有统计学意义(P<0.05),低、中、高剂量爱维治组小肠黏膜上皮细胞中IL-2和IL-6水平高于正常组,但差异无统计学意义(P>0.05)。爱维治各组大鼠小肠绒毛高度、黏膜厚度、隐窝深度、全层厚度与放射组比较改善显著(P<0.05)。而中、高剂量爱维治组小肠上皮黏膜细胞中凋亡相关蛋白bax表达水平显著低于放射组,而bcl-2水平高于放射组,差异有统计学意义(P<0.05)。结论爱维治能有效抑制凋亡蛋白bax水平的表达,并可抑制凋亡蛋白bal-2表达,对放射性肠炎上皮黏膜细胞具有良好的修复效果。Objective To investigate the effect of radiation on secretion of small intestinal epithelial cells IL-2 and IL-6, and deproteinized calf blood extract (Actovegin) on the inhibition of apoptosis of small intestinal epithelial cells. Methods 50 rats were randomly divided into normal group, radiation group,low-dose Actovegin group, medium-dose Actovegin group and high- dose Actovegin group,with 10 rats in each group. High-energy X-ray linear accelerator was used on radiation group,low-, medium- and high-dose Actovegin group, the dose was 9.0 Gy, and the rat radiation damage model was established. Modeling rats were in- jected drug in abdomen for 4 days, pathological section of small intestinal epithelial cells was taken, IL-2 and IL-6 level was detec- ted,and effect of Actovegin on small intestinal epithelial cells apoptosis was detected by immunohistocbemistry. Results Small intestinal epithelial cells IL-2 andIL-6 level of radiation group was significantly higher than the other groups, the difference was statistically significant (P 〈 0.05 ), L-2 andIL-6 level of low-, medium-and high-dose Actovegin group was higher than the normal group, but the difference was not statistically significant ( P 〉 0.05 ). Intestinal villus height, mucosal thickness, crypt depth, full thickness of Actovegin group improved significantly compared to the radiation group (P 〈 0.05 ). Intestinal epithelia| mucosal ap- optosis-related proteins Bax expression level of the high-dose Actovegin group was significantly lower than the radiation group, bcl- 2 level was higher than the radiation group, the difference was statistically significant (P 〈 0.05). Conclusion Actovegin can ef- fectively inhibit the expression of the apoptotic proteins Bax, and may contribute to the inhibition of apoptosis protein bal-2 expres- sion, It has ~ood repair effect.
