大豆异黄酮对内毒素及D-氨基半乳糖所致小鼠急性肝损伤的保护作用

摘  要:为研究大豆异黄酮对D-氨基半乳糖(GalN)和内毒素(LPS)联合所致小鼠急性肝损伤的保护作用,将试验小鼠按体重随机分为5组,即正常组、模型组、水飞蓟素组(阳性对照组)及大豆异黄酮高、低剂量组。每日给药1次,连续7 d。除正常组外,其余小鼠腹腔注射GalN及LPS建立急性肝损伤模型,苏木素-伊红(HE)染色法观察肝组织病理学变化,ELISA法检测血清肿瘤坏死因子-α(TNF-α),比色法检测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)以及肝脏一氧化氮(NO)、过氧化脂质(LOOH)、丙二醛(MDA)含量和Caspase-3、Caspase-8活化水平。结果表明:大豆异黄酮显著降低GalN及LPS联合诱发的急性肝损伤小鼠血清ALT、AST活性,减轻肝组织病理损伤,降低血清TNF-α和肝NO水平,降低肝LOOH和MDA水平,降低肝组织Caspase-3和Caspase-8活化水平。说明大豆异黄酮对GalN和LPS联合诱导的小鼠急性肝损伤具有保护作用,其机制可能与其抗炎、抗氧化活性和抗肝细胞凋亡作用有关。For the purpose of studying the protective effect of soy isoflavones on acute liver injury induced by D-galactosamine ( GaIN ) and lipopolysaeeharide (LPS) in mice, the tested mice were randomly assigned to 5 groups : normal control, model control,silymarin( positive control) ,as well as high and low dose groups of soy isoflavones. Animals were treated once daily for 7 days. GaIN and LPS were given intraperitoneally to the mice of groups, then the pathological changes was detected with hematoxylin & eosin( HE )staining, tumor necrosis factor-α (TNF-ct)was detected by ELISA method, and the alanine aminotransferase ( ALT), aspartate aminotransferase ( AST ), lipid hydroperoxide ( LOOH ), malondialdehyde ( MDA ), nitric oxide ( NO ), and the activation of Caspase-3 and Caspase-8 were detected by the colorimetric method. The results showed that the soy isoflavones reduced the serum ALT and AST, alleviated the hepatic lesions induced by GaIN and LPS, decreased serum TNF-α and hepatic NO levels,reduced the hepatic LOOH and MDA contents, and decreased the Caspase-3 and Caspase-8 activations of liver tissues. It is suggested that soy isoflavones have protective effect on acute liver injury induced by GaIN and LPS in mice, probably via the anti-inflammatory, antioxidative and anti-apoptotic ability.
