摘  要:目的探讨银杏叶片对大鼠肝纤维化的疗效及部分可能机制。方法将造模成功后的20只WISTAR大鼠随机分为治疗组及自然恢复组,治疗组正常进食水且每日以银杏叶片灌胃,空白对照组及自然恢复组正常进食水,同时每日以等量自来水灌胃。2个月后留取血清、肝组织,测定其血清透明质酸(HA)含量及肝组织中羟脯氨酸(Hpy)含量,行组织病理学检查评价肝纤维化形成情况,并行免疫组化检查观察过氧化物酶体增殖物激活受体(PPAR-γ)的表达情况。结果自然恢复组及治疗组大鼠血清HA及肝组织Hpy浓度与空白对照组相比较均升高(P<0.05),其中自然恢复组较治疗组升高明显(P<0.05)。免疫组化结果显示PPAR-γ在空白对照组表达最显著,自然恢复组及治疗组较空白对照组表达均下调(P<0.05),而与自然恢复组相比,治疗组大鼠肝脏PPAR-γ表达水平明显上调(P<0.05)。结论银杏制剂治疗肝纤维化的机制之一可能为上调PPAR-γ水平的表达。Objective To investigate the therapeutic effect of ginkgo leaves on hepatic fibrosis so as to find a theoretical basis for the clinical aplication of ginkgo leaves preparation. Methods Fifty male Wistar rats were randomly assigned into normal control group (n=10) and model group (n=40). All the rats in model group were subcutaneously given CCL4 injection, high-fat and low-protein diet and alcohol to establish hepatic fibrosis models. 8 week later three rats were sacrificed to make sure the success of hepatic fibrosis model. The survived 20 rats in model group were again randomly divided in to 2 groups which were spontaneous recovery group and treated group. Rats in treated-group were conventionally fed and perfused with extracts of ginkgo leaves every day, while the rats in normal con- trol group and spontaneous group were conventionally fed and per{used with water. 8 weeks later, the liver index, levels of serum hyaluronic acid (HA) and liver hydroxyproline (Hyp) were measured, and the degree of hepatic fibrosis was estimated. The expression of peroxisome proliferator-activated re- ceptors γ (PPAR-γ) was detected by immunohistochemistry. Results Compared with normal control group, the serum HA and the liver Hyp increased in spontaneous recover group and the treated group (P〈0.05) , while the serum HA and the liver Hyp in treated group decreased compared with sponta- neous recover group (P〈0.05). The expression of PPAR-γ was highest in normal control group. Compared with normal control group, the expression of PPAR-γ decreased in spontaneous recovergroup and the treated group(P〈0.05), while compared with spontaneous recover group, the expres- sion of PPAR-γ in treated group increased remark(P〈0.05). Conclusion The extracts of ginkgo may palliate the hepatic fibrosis by regulating the expression of PPAR-γ.
