摘 要:[目的]研究VNTR基因座D1S80在朝鲜族和汉族群体中的遗传多态性.[方法]采用PCR扩增结合聚丙烯酰胺凝胶电泳技术和银染法对207名无关朝鲜族个体和203名无关汉族个体的D1S80基因座进行群体遗传多态性调查.[结果]D1S80基因座在朝鲜族和汉族人群中共检出26个等位基因,共103种基因型,朝鲜族群体中检出25个等位基因,73种基因型;汉族群体中检出22个等位基因,71种基因型;两个群体皆符合Hardy-Weinberg平衡,朝鲜族群体的杂合度观察值、多态性信息量、非父排除率和个人识别能力分别为0.917 9,0.893 8,0.705 9,0.968 3,汉族群体中分别为0.803 0,0.870 3,0.684 2,0.968 8;将两个群体的等位基因分布频率分别与其他不同地区、民族、种族群体进行比较,结果证实D1S80基因座在不同地区、民族和种族之间存在着不同程度的差异.[结论]D1S80基因座在朝鲜族和汉族群体中具有高度多态性,可用于法医学个体识别、亲权鉴定和群体遗传学调查.OBJECTIVE To study the genetic polymorphism of the variable number of tandem repeat (VNTR) locus DIS80 in Korean-Chinese and Han nationality population. METHODS By using PCR, polyacrylamide gel electrophoresis and silver staining, the population genetic polymorphism of locus D1S80 was surveyed in 207 of unrelated Korean-Chinese and 207 of unrelated Han nationality. RESULTS 26 alleles and 103 kinds of genotypes were detected in both two groups, including 25 alleles and 73 kinds of genotypes in Korean-Chinese group, and 22 alleles and 71 kinds of genotypes in Han nationality group. Both of them were The accorded with Hardy Weinberg hetrozygosity observation, equilibrium. hetrozygosity expections, polymorphism information content, probability of exclution and discrimination power were 0. 917 9, 0. 881 7, 0. 893 8, 0. 705 9, 0. 968 3 in Korean-Chinese group, and 0. 803 0, 0. 873 0, 0. 870 3, 0. 684 2, 0. 968 8 in Hart nationality group, respectively. Moreover, the study on the comparation between this two groups and different ethnic in different areas confirmed that it had differences to varying degrees among different ethnic in different areas. CONCLUSION D1S80 loci is a highly polymorphic VNTR loci and valuable in individual identification and paternity identification.
