摘  要:目的:研究重组组织型纤溶酶原激活剂(rt-PA)对大鼠脑梗死溶栓治疗后脑组织中ZO-1蛋白的表达,探讨不同剂量rt-PA对脑梗死溶栓后血脑屏障的影响。方法:采用大鼠自体血栓栓塞法制备大脑中动脉闭塞动物模型,采用免疫荧光染色技术检测ZO-1蛋白的表达水平,以及比色法测定血脑屏障对伊文思蓝(evans blue,EB)的通透率。结果:与正常对照组及假手术组比较,低剂量rt-PA在溶栓治疗后脑组织中ZO-1蛋白表达升高,而且EB含量显著减低;而高剂量rt-PA组ZO-1表达降低,脑组织中EB含量显著升高。结论:低剂量rt-PA在大鼠脑梗死溶栓治疗后可能对血脑屏障存在保护作用。Objective To study the expression of ZO-1 protein in the brain tissue after treatment with recombinant tissue-type plasminogen activator (rt-PA) on rats with cerebral infarction ard to explore the effects of different doses of rt-PA on the blood-brain barrier. Methods Rat models of the middle cerebral artery occlusion were prepared with autologous thromboembolism, and ZO-1 protein level was detected by immune-fluore scence staining. Besides, colorimetric technique was applied to evaluate the penetration rate of Iraq Evans Blue (Evans Blue, EB) through the blood-brain barrier. Results By comparing with the normal control group and the sham group, the expression of ZO-1 protein in brain tissues increased and EB reduced significantly while low-dose of rt-PA was applied in the thrombolytic therapy; it showed opposite results while high doses of rt-PA were applied Conclusion Low dose of rt-PA maybe have protective effect on the blood-brain barrier after thrombolytic therapy in rats with cerebral infarction.
