肝硬化大鼠胃粘膜损害的研究

摘　　要：目的探讨Toll样受体4(TLR4)介导的信号传导在肝硬化大鼠模型胃粘膜损害中的作用。方法将16只雌性SD大鼠随机分为对照组和肝硬化组,每组8只,肝硬化组大鼠自由饮用0.01%的二乙基亚硝胺(四川省农业科学研究院农药研究所提供)2个月。用荧光定量逆转录-聚合酶链反应(RT-PCR)对大鼠胃粘膜组织细胞中Toll样受体4(TLR4)和肿瘤坏死因子α(TNF-α)mRNA做定量分析。结果肝硬化组TLR4和TNF-αmRNA的表达高于对照组(P<0.05)。结论大鼠肝硬化时,TLR4介导的信号传导在胃粘膜损害中起了重要作用。Objective To study the role of Toll-like receptor4（TLR4） mediated signal transduction in the damage of gastric mucosa with liver cirrhosis rat model.Methods 16 SD female rats were randomly assigned to two groups：control group and cirrhosis group,there were 8 rats in each group.The rats in cirrhosis group drinking the water with 0.01% diethylnitrosamine in two Months to form the rat model of liver cirrhosis.Real-time reverse transcription-polymerase chain reaction（RT-PCR） was used to quantitatively determine the expressions of TLR4、TNF-α mRNA in gastric mucosa tissues.Results The expression of TLR4 and TNF-α mRNA in cirrhosis group was significant higher than control group（P0.05）.Conclusion The TLR4 mediated sig nal transduction play a part in the damage of gastric mucosa with liver cirrhosis rat model.
