摘  要:目的探讨葡萄籽提取物原花青素(grape seed procyanidin extract,GSPE)对人喉表皮样癌Hep-2细胞增殖和凋亡的影响。方法 Hep-2细胞与25、50、100和200μg/mL的GSPE共同孵育48 h后,采用噻唑蓝(MTT)法检测细胞增殖抑制率,流式细胞术分析细胞凋亡情况;用Western blot法检测不同浓度的GSPE作用于肿瘤细胞时,凋亡相关蛋白Bcl-2,bax表达情况。结果不同浓度(25、50、100和200μg/mL)GSPE对Hep-2细胞的增殖抑制率分别为(20.0±2.3)%、(33.6±2.7)%、(52.7±2.1)%和(76.6±2.3)%;凋亡率分别为(4.99±0.02)%、(26.49±2.49)%、(46.09±1.18)%和(58.77±2.38)%;细胞增殖抑制率和凋亡率均随GSPE浓度的升高而增高(P<0.05);Western blot检测药物处理细胞后Bcl-2表达水平下调,而Bax表达水平明显上调。结论 GSPE在体外可呈浓度依赖性抑制人喉表皮样癌Hep-2细胞增殖并促进其凋亡。Objective To investigate the effects of grape seed procyanidin extract (GSPE)on proliferation and apoptosis of human laryngeal carcinoma cell line Hep-2. Methods Hep-2 cells were cultured with different concentrations of GSPE (25, 50, 100 or 200 μg/mL)for 48 hours, inhibition of cell proliferation in vitro was measured by MTT assay, and apoptosis was detected by flow cytometry ; The expression of the Bcl-2, bax which associated with cell apoptosis were measured by Western blot. Results The inhibition rates were (20. 0± 2. 3 )%, (33.6 ±2. 7)%, ( 52. 7 ±2. 1 )% and (76. 6 ±2. 3)%, respectively, while the apoptotic rates were (4.99 ±0.02)%, (26.49±2.49)%, (46.09±1.18)% and (58.77±2.38)%. Both the inhibition rate and the apoptotic rate elevated with the increasing GSPE concentration ( P 〈 0.05 ) ; Western blot also showed that GSPE down-regulated Bcl-2 and up-regulated Bax. Conclusion GSPE inhibits the growth of Hep-2 cells and induces apoptosis in vitro in a concentration-dependent manner.
