摘　　要：目的　通过对人肝癌细胞株 (SMMC 772 1)转染TGF β1基因 ,观察该细胞MMP 2 ,9表达的变化。方法　用电穿孔法进行基因转染 ,并经Westernblot ,Northernblot法作鉴定 ,后分别应用Northernblot,Westernblot法观察该细胞株表达MMP 2 ,9mRNA和MMP 2蛋白的变化。结果　经转染TGF β1基因的细胞株MMP 2mRNA和蛋白表达均高于对照组 ,但MMP 9mRNA表达则没有变化。结论　TGF β1可促进肝癌细胞MMP 2表达 ,表明TGFPurpose To study the expression of MMP?2,9 on human hepatoma cells(SMMC?7721)transfected with TGF?β 1 gene. Methods TGF?β 1 gene transfection was done by electroporation.The expression of TGF?β 1 and MMP?2,9 on transfectants was detected by Northern blot and Western blot analysis. Results Higher mRNA and protein level of MMP?2 were observed in SMMC?7721 cells transfected with TGF?β 1 gene,however,the expression of MMP?9 was not affected by TGF?β 1. Conclusions TGF?β 1 might upregulate the expression of MMP?2 on human hepatoma cells,it is possible that TGF?β 1 is involved in the invasion and metastasis of human hepatoma.
