摘 要:研究二次低能离子束诱变对石油污染土壤中微生物原油降解能力及产表面活性剂能力的影响。选取经一次低能粒子束诱变筛选出的高效产表活烃降解菌23A作为出发菌株,对其进行第二次低能离子束注入诱变。二次诱变后可以产生降解原油效果更好的菌株,并且能降低表面张力,所筛选出的菌23A-3,对原油的降解率比出发菌高17.3%,产生的表面活性物质使表面张力低于出发菌25%。结果表明:利用低能离子束对烃降解微生物进行二次诱变改良是可行的,对烃降解菌降解水平的进一步提高具有现实意义,同时也丰富了离子束生物工程学的技术路线。To study the effects of secondary mutation on the ability for degradation of petroleum hydrocarbon and producing biosurfactant, microorganism from the oil contaminated soil was implanted again with low-energy N+ ions. Mutant strain 23A, obtained after the primary mutation with the low-energy N+ which could efficiently degrade oil and produce biosurfactant, was implanted again with low-energy N+. Secondary mutant strain degrades oil better and reduces the surface tension of culture medium significantly. Compared with the original strain 23A, mutant strain 23A-3 increases the degradation rate of total petroleum hydrocarbon by 17.3% and decreases the surface tension of culture medium by 25%. The results indicate that the secondary mutation has great potential to gain more effective microorganism. It has practical significance to improve the level of petroleum degradation further, and can enriche the technical route of ion beam bioengineering.
