Optimization of Ultrasonic Extraction Technology for Zanthoxylum nitidum by Orthogonal Design and in Dynamic Process

摘  要:以总生物碱提取率为指标,先用正交试验优化两面针的超声提取工艺,再动态过程精选提取工艺。最佳工艺条件为:复合酶预处理后,以体积分数60%乙醇(盐酸5 g/L)超声(250 W)提取3次,第1次以10倍量溶剂提取15 min,第2次以4倍量溶剂提取12 min,第3次以3倍量溶剂提取9 min,总生物碱提取率87.80%。该工艺高效、节能、省时,为工业生产奠定了实验基础。The ultrasonic extraction technology for Zanthoxylum nitidum was optimized by orthogonal test at first, and fur- ther optimized in dynamic process ,with extraction rate of total alkaloids as the index. The optimal process was as follow- ing:Z, nitidum powder was hydrolyzed by ceUulase of 1:250 and pectinase of 1:250 in 4 times buffer with pH 5 for 30 rain at ambient temperature (30 ~C ) ,and then was extracted 3 times by ultrasonic wave (250 W) with 60% ethanol (hydrochloric acid 5 g/L) as solvent,extracted 15 rain with 10 times solvent at the first time,extracted 12 min with 4 times solvent at the second time, extracted 9 rain with 3 times solvent at the third time. The extraction rate of total alka- loids was 87.80%. The process is efficient,energy and time saving and can be used as the experimental base of industri- al production.
