摘 要:应用表面等离子共振(SPR)技术检测过敏性休克死亡豚鼠血清类胰蛋白酶,试图为过敏性休克死亡提供客观诊断依据。首先,将16只健康豚鼠随机均分为实验组和对照组,每组再分为死亡即时组和冷冻3 d组。以人混合血清诱发动物过敏性休克致死。采用分子自组装方法将抗类胰蛋白酶单克隆抗体(Anti-TPSAB1)固定在SPR生物传感器芯片表面,检测豚鼠血清中类胰蛋白酶。实验结果表明:实验组类胰蛋白酶SPR响应值比对照组显著升高,在冷冻3 d的条件下对豚鼠血清类胰蛋白酶的表达无明显影响。因此,基于SPR生物传感器在动物血清类胰蛋白酶方面具有很好的应用前景,可作为诊断过敏性休克死亡的一项依据。Surface plasmon resonance(SPR)technology is used to detect serum tryptase levels in guinea pigs died from anaphylactic shock.The aim of this study is attempted to provide an objective evidence for diagnosis of anaphylactic shock.Firstly,sixteen healthy guinea pigs are randomly divided into experimental group and control group.For each group animals are sub-grouped into animals of instantaneous death and animals kept at-20 ℃ for three days freezing.Anaphylactic shock is induced by injection of human pooled serum.Anti-TPSAB1 are immobilized on the surface of SPR biosensor chips using self-assembly method,and then SPR is adopted to detect the tryptase in guinea pigs serum.The experiments show that the response value of experimental group of tryptase is significantly higher than the control group.The three days freezing condition does not affect the expression for serum tryptase.Therefore biosensor based on SPR has good application prospect in detection of animal serum tryptase,and expression of detection of tryptase may act as an evidence for diagnosis of anaphylactic shock.
