摘  要:目的探讨津力达口服液对高脂饮食负荷的2型糖尿病大鼠体内氧化应激标志物的影响。方法将40只SD雄性2型糖尿病模型大鼠随机分为糖尿病模型组和津力达治疗组,每组20只,另选20只正常SD雄性大鼠作为正常对照组。津力达治疗组给予津力达口服液5 ml/kg灌胃、2/d,正常对照组及糖尿病模型组给予同剂量0.5%羟甲纤维素钠溶液灌胃。8周后处死大鼠,测定体质量、血糖、甘油三酯(TG)、总胆固醇(TC)、肾小球截面积及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物歧化酶(GSH-Px)、丙二醛(MDA)、高级糖化终产物(AGE)等肾脏氧化应激指标。分别在给药第4、8周末剪鼠尾采血,测定血SOD、GSH-Px、MDA、一氧化氮(NO)、一氧化氮合酶(NOS)及AGE等指标。结果给药第8周末,津力达治疗组、糖尿病模型组与正常对照组相比体质量均明显偏低(P<0.05);津力达治疗组空腹血糖、TC、TG均明显低于糖尿病模型组(P<0.05),除空腹血糖(P<0.05)外,其余指标与正常对照组比较差异无统计学意义(P>0.05)。津力达治疗组与糖尿病模型组比较,肾小球截面积减小(P<0.05),肾组织AGE、MDA降低(P<0.05),SOD、GSH-Px升高(P<0.05);给药第4、8周,津力达治疗组血SOD、GSH-Px、NO、NOS高于糖尿病模型组(P<0.05),MDA、AGE-P低于糖尿病模型组(P<0.05)。给药第8周末,糖尿病模型组、津力达治疗组GSH-Px与本组第4周末比较显著升高(P<0.05)。结论津力达口服液能有效改善高脂饮食负荷的2型糖尿病大鼠体内氧化应激标志物的表达。Objective To investigate influences of Jinlida oral liquid on markers of oxidative stress and antioxi- dant reserve in rats with type 2 diabetes mellitus. Methods A total of 40 male Sprague-Dawley (SD) rats with type 2 diabetes mellitus were randomly divided into diabetes model group ( group A, n = 20) and Jinlida therapy group ( group B, n = 20) , and other 20 normal male SD rats were recruited as control group. Group B was treated with 5 ml/kg lavage orally (2 times/d) , while group A and control group were given the same dose of 0.5 % sodium earboxmethylcellulose so- lution. After weeks, rats were sacrificed, and body weight, blood glucose, triglyceride (TG) , total cholesterol (TC), glomerular cross-sectional area and superoxide dismutase (SOD) , glutathione peroxidase (GSH-Px) , malondialdehyde (MDA) and advanced glycosylation end product were detected. At the end of 4th and 8th week, SOD, GSH-Px, MDA , nitric oxide (NO), nitric oxide synthase (NOS) and AGE in blood of rats' tails were detected respectively. Results Body weight in group A and group B were significantly lower than that in control group at the end of 8th week ( P 〈 0.05 ) ; Levels of fasting blood glucose, TC and TG in group B were significantly lower than those in group A (P 〈 0.05 ) , and there was no statistically significance in indexes compared with those in control group (P 〉 0.05) , except the fasting blood glucose index (P 〈 0.05). The glomerular cross-sectional area, AGE and MDA in nephridial tissue in group B were significantly lower than those in group A (P 〈 0.05 ) , while activities of SOD and GSH-Px in renal tissue were sig- nificantly higher (P 〈 0. 05). At medication for 4 weeks and 8 weeks, serum concentrations of SOD, GSH-Px and NO, and NOS in group B were higher than those in group A (P 〈 0.05 ) , and levels of MDA and AGE-P were lower than thosein group A (P 〈 0.05 ).
