摘 要:目的研究双歧杆菌黏附素对应激大鼠肠黏膜炎性反应的影响。方法将48只实验大鼠随机分成应激组、应激+黏附素组,每组24只,以束缚为应激条件,在应激组和应激+黏附素组大鼠建立应激模型,造模后进入实验期,实验8 d。两组大鼠每日给予相同热量、氮量(热量125.4 kJ/kg,氮量0.2 g/kg)的肠内营养,应激+黏附素组大鼠每日喂养肠内营养加双歧杆菌黏附素(5 mg/kg),应激组大鼠每日喂养肠内营养加等量的生理盐水(5 mg/kg),测定造模前、造模后、实验第3天和第8天两组大鼠肠黏膜和血浆白细胞介素-6(IL-6)、白细胞介素-10(IL-10)、肿瘤坏死因子-α(TNF-α)、干扰素-γ(IFN-γ)。结果与造模前比较,造模后应激组和应激+黏附素组肠黏膜和大鼠血浆IL-6、TNF-α、IFN-γ水平浓度明显升高;实验第3天和第8天,应激组大鼠肠黏膜和血浆IL-6、IFN-γ、TNF-α明显高于造模前。与造模后和应激组比较,实验第3天和第8天,应激+黏附素组大鼠肠黏膜和血浆TNF-α、IL-6和IFN-γ水平明显降低,而IL-10水平明显升高。结论双歧杆菌黏附素可下调肠黏膜炎性反应,减轻肠黏膜应激损伤。Objective To investigate the effect and mechanism of bifidobacterial adhesin on the intestinal barrier function in stress rats.Methods Forty-eight rats were randomly divided into stress group(n=24)and stress+bifidobacterial adhesin group(n=24).Taking fettering as the stress condition,the model of stress was established on rats in stress group and stress+bifidobacterial adhesin group,and then began the experimental period which lasted 8 days.Both groups of rats were given enteral nutrition with equal heat,nitrogen(heat 125.4kJ/kg,nitrogen 0.2g/kg).Stress+bifidobacterial adhesin group of rats was fed with enteral nutrition plus 5mg/kg bifidobacterial adhesin.Stress group of rats was fed with enteral nutrition plus equivalent normal saline(5mg/kg).The plasma levels of interleukin-6(IL-6),interleukin-10(IL-10),tumor necrosis factor(TNF-α),interferon-γ(IFN-γ),and the level of IL-6,TNF-α and IFN-γ in intestinal mucosal were measured.Results After modeling,the levels of IL-6,TNF-α,and IFN-γ were significantly higher in both groups than the levels before modeling in intestinal mucosal and plasma.The levels of IL-6,IFN-γ,TNF-αin plasma and intestinal mucosal were significantly higher in stress group than the levels before modeling on the 3days and the 8days.On the third day and the eighth day,the plasma levels of IL-6,IFN-γand TNF-α,also the levels in intestinal mucosal were significantly lower in stress+bifidobacterial adhesin group than the levels in stress group and after modeling,and the levels of IL-10 were significantly higher.Conclusion Bifidobacterial adhesin may reduce the inflammatory response and stress injury,thus provides protective effects on intestinal mucosalintestinal in stress rats.
