摘 要:目的:探讨血清应答因子(SRF)、E-钙黏蛋白(E-cadherin)和β-连环蛋白(β-catenin)在肝细胞癌中的表达及其与临床病理特征和预后的关系。方法:运用组织芯片技术,通过免疫组织化学EnVision法检测182例肝细胞癌和92例癌旁肝组织中SRF、E-cadherin和β-catenin的表达情况,并分析其与临床病理特征和预后的关系。结果:SRF在肝细胞癌中阳性表达率为75.8%,明显高于癌旁肝组织(为21.7%),差异具有统计学意义(P<0.05);肝细胞癌E-cadherin和β-catenin阳性表达率分别为59.3%和58.8%,亦高于癌旁肝组织(分别为30.4%和26.1%),差异具有统计学意义(P<0.05)。肝细胞癌中SRF的表达与TNM分期、转移和术后复发相关(P<0.05);E-cadherin和β-catenin异常表达与脉管内瘤栓、TNM分期、转移和术后复发相关(P<0.05)。肝细胞癌中SRF表达与E-cadherin、β-catenin异常表达呈正相关。结论:肝细胞癌中SRF的高表达,与E-cadherin和β-catenin表达的下调,可能是肝细胞癌侵袭和转移的重要机制之一,在肝细胞癌发生、发展中起到重要作用。Objective: To investigate the expression of serum response factor (SRF), E-cadherin and β-catenin in hepatocellular carcinoma and its relation to clinicopathological characterisitics of the tumor and prognosis. Methods: Tissue microarray and immunohistochemical study (EnVision method) for SRF, E-cadherin and β- catenin was performed on tissue microarray which consisted of 182 cases of hepatocellular carcinoma, 92 cases of adjacent non-neoplastic liver tissues. Results: In the hepatocellular carcinoma group 75.8% tumor samples expressed SRF, and the levels is significantly higher than that in surrounding liver tissue (21.7%, P〈0.05). The expression of SRF was positively correlated with the TNM stage, tumor metastasis and recurrence (P〈0.05). In the hepatocellular carcinoma group, aberrant expression of E-cadherin andβ-catenin was 59.3% and 58.8% respectively, and these levels were higher than those in the adjacent non-neoplastic liver tissue group (30.4% and 26.1% P〈0.05). Aberrant expression of E-cadherin and β-catenin was related to vascular cancer embolus, the TNM stage, tumor metastasis and recurrence (P〈0.05). The expression of SRF correlated with aberrant expression of E-cadherin and [3-catenin in hepatocellular carcinoma. Conclusion: In hepatocellular carcinoma, the overexpression of SRF and the downregulated expression of E-cadherin andβ--catenin may be important mecha- nisms for promoting the invasion and metastasis of hepatocellular carcinoma and may also play a key role in the occurrence and development of hepatocellular carcinoma.
