细胞周期相关蛋白CyclinDl、CyclinE、p16和p21在卵巢高级别浆液性癌中的表达及其意义

摘　　要：目的:检测卵巢高级别浆液性癌中细胞周期蛋白(Cyclin)D1、CyclinE及其抑制因子p16、p21的表达,探讨它们在卵巢肿瘤发生发展中的作用以及与临床病理特征的关系。方法:收集卵巢高级别浆液性癌50例,卵巢交界性浆液性囊腺瘤33例,制作石蜡组织芯片,免疫组织化学染色,分析其CyclinD1、CyclinE、p16、p21表达情况及临床病理联系。结果:CyclinD1、p21在交界性浆液性囊腺瘤中的表达与高级别浆液性癌比较,差异均无统计学意义,而CyclinE、p16在交界性浆液性囊腺瘤中的表达均低于高级别浆液性癌,差异有统计学意义。四种蛋白的表达与卵巢高级别浆液性癌患者年龄、临床分期、淋巴结转移、腹水均无显著相关。结论:卵巢交界性浆液性囊腺瘤和高级别浆液性癌的细胞周期的调控机制可能不同,CyclinE的过表达在卵巢高级别浆液性癌的发病中的作用较CyclinD1重要;细胞周期调控的紊乱在卵巢高级别浆液性癌的发生和发展过程中起了重要的作用。Objective： To detect the expression of CyclinD 1, CyclinE and their inhibitors p16, p21 in highgrade ovarian serious adenocarcinoma and their clinical significance. Methods： The paraffin-embedded tissues of 33 cases of ovarian serous borderline tumor and 50 cases of ovarian high-grade serious adenocarcinoma were collected and made into tissue microarray. Expression of of CyclinD1, CyclinE, pl6 and p21 were detected with immunohistochemistry. And their clinicopathological significance was analyzed. Results： There was no signifi- cant difference between the expressions of CyclinD1 and p21 in ovarian serous borderline tumor and those in high- grade serious adenocarcinoma, while the expressions of CyclinE and pl6 were lower than those in high-grade serious adenocarcinoma. There were no correlation among the expressions of four proteins and patients＇ age, clinical stages, lymph node metastasis and ascites. Conelusion： The regulations of cell cycle in ovarian serous borderline tumor and high-grade serious adenocarcinoma are different. CyclinE may play a more important role in the pathogenesis than CyclinD 1 does. The deregulation of cell cycle controlling has significant importance in the develolament of ovarian high-grade serious adenocarcinoma.
