多囊卵巢综合征大鼠卵巢smad3的表达及其组织病变的观察

摘  要:目的定量分析卵巢血管密度,探讨多囊卵巢综合征(PCOS)大鼠卵巢smad3的表达与体积增大、微血管密度增加等病理改变的相关性。方法皮下注射脱氢表雄酮法建立PCOS大鼠病理模型,动情间期取卵巢组织后称重;免疫组织化学法检测smad3在卵巢组织中表达,anti-CD34标记卵巢微血管,并进行定量分析;蛋白免疫印迹法(Western blot)检测卵巢smad3的表达;逆转录聚合酶连反应(RT-PCR)法检测smad3 mRNA在卵巢组织中的表达。结果免疫组织化学检测显示:与对照组比较,实验组卵泡smad3表达减弱,卵巢间质表达增强(P<0.05),在各级卵泡及间质均有表达。实验组卵巢体积增大,卵巢间质微血管的密度增加(P<0.05);Western blot检测结果提示在实验组smad3表达上调;RT-PCR结果显示实验组卵巢组织中smad3 mRNA表达与对照组无显著性差异(P>0.05)。结论 PCOS卵巢间质平均血管密度增加,smad3的表达变化可能与PCOS特殊卵巢病理改变有关。Objective: To investigate the changes of smad3 expression and microvessel density in ovary and analyze their correlation with pathological change in rats with polycystic ovary syndrome (PCOS). Methods: The rats with PCOS were established by injecting subcutaneously dehydroepiandrosterone (DHEA). The weight of ovaries was determined in anestrum. The detection of smad3 protein was carried out by immunohistochemistry and Western blot. The capillary vessels were marked by anti-CD34 and evaluated quantitatively. The reverse transcription polymerase chain reaction (RT-PCR) was employed to analyze the transcriptional level of smad3 mRNA. Results: The results of immunohistochemistry showed that smad3 was lowly expressed in the follicles,but increased in the stroma of ovary of rats with PCOS (P〈0.05). The ovary volume of experimental rats was increased compared with control rats. The microvessel density of ovaries stroma in rats with PCOS was higher than that in control rats. The significant difference of smad3 mRNA in ovaries between the two groups was not found. Conclusions: The microvessel density of ovaries stroma in rats with PCOS is increased. The change of smad3 expression may contribute to pathological appearance of ovaries in rats with PCOS.
