摘 要:目的建立同时测定宣木瓜药材中五种有机酸(莽草酸、苹果酸、柠檬酸、绿原酸、咖啡酸)和原儿茶酸的含量测定方法。方法采用HPLC法,Phenomenex luna C18色谱柱(250 mm×4.6 mm,5μm);流动相A:0.5%磷酸二氢铵溶液(磷酸调节pH值至2.40);流动相B:乙腈进行梯度洗脱;检测波长:214 nm;流速:1.0 mL·min-1;进样量:10μL;柱温:30℃。结果莽草酸、苹果酸、柠檬酸、绿原酸、咖啡酸五种有机酸和原儿茶酸的线性范围分别为:0.049 5~0.371 g·L-1(r=1.000 0),0.066 3~0.358g·L-1(r=0.988 9),0.067 5~0.352 g·L-1(r=0.995 7),0.009 9~0.080 2 g·L-1(r=0.991 0),0.008 81~0.062 2 g·L-1(r=0.992 1),0.010 1~0.090 2 g·L-1(r=0.993 3),平均加样回收率分别为:97.2%(RSD=2.0%)、99.6%(RSD=4.1%)、101.0%(RSD=1.6%)、98.4%(RSD=2.3%)、99.5%(RSD=2.1%)、101.0%(RSD=1.4%)。结论该实验建立了同时测定木瓜中五种有机酸和原儿茶酸的HPLC检测方法,操作简便,可靠,灵敏度较高,具有较好的重复性和稳定性,可用于宣木瓜药材的质量控制。Objective To establish an HPLC method for the determination of five kinds of organic acids ( shikimic acid, citric acid, malic acid,chlorogenic acid,caffeic acid) and protocatechuic acid in Chaenomelis Fructus. Methods A high performance liquid chromatogra-phy equipped a Phenomenex luna C lS (250 mm ×4.6 mm,5 μm) column with UV detection was used . The mobil phase consisited of 0.5% ammonium dihydrogen phosphate aqueous solution (A) and acetonitirle (B) with gradient elution. The detection wavelength was set at 214 nm and the flow rate was 1.0 mL · min^-1. The injection volumn was 10 μ1 and the column temperature was 30℃. Results The linear ranges of shikimic acid,citric acid,malic acid,chlorogenic acid,caffeic acid and protocatechuic acid were 0.049 5 -0.371 g·L^-1(r=l.000 0) ,0.066 3 - 0.358 g·L^-1(r= 0.988 9) ,0.067 5 - 0.352 g·L^-1(r=0.995 7) ,0.009 90-0.080 2 g·L^-1(r = 0.991 0) ,0. 008 81 - 0. 062 2 g·L^-1 ( r = 0. 992 1 ), 0.010 1 - 0. 090 2 g·L^-1 ( r = 0. 993 3 ) respectively. The average recoveries were 97. 2% ( RSD = 2. 0% ) ,99. 6% ( RSD =4.1%),101.0%(RSD =1.6%),98.4%(RSD =2.3%),99.5%(RSD =2.1%), 101.0% ( RSD = 1.4% ) respectively. Conehlsions This test establishes a method that can detect five kinds of organic acids and proto-catechuic acid in Chaenomelis Fructus. The method is simple, convenient, reliable and has a high sensitivity as well as a good reproduc-ibility and stability,which can be used for quality control of Fructus Chaenomelis.
