摘 要:从富硒土壤中筛选富硒微生物、并进行富硒条件优化和分子鉴定。选取加硒质量浓度、加硒时间和培养温度3个因子,设计富硒条件优化正交试验。用原子荧光法测定菌体中硒含量,得到其富硒的最优条件为Na2SeO3溶液质量浓度20μg/mL,加硒时间5h后,培养温度37℃。在此条件下培养菌体硒含量平均为0.7108μg/mL,富硒率为58.3%。经鉴定该富硒微生物为一株枯草芽孢杆菌。This experiment focused on the screening, optimization of Se-enriched condition and identification of selenium-enriched microbe. To maxi- mize its selenium-enriched ability, parameters such as the selenium concentration, the time of adding NazSeO3 and the incubation temperature were investigated by orthogonal array design. The results showed that the maximized quantity was obtained by culturing the microbe for 5h at 37℃ and adding Na2SeO3 solution by the concentration of 20μg/ml. Under these conditions, the Se-enriching quantity was 0.7108μg/ml, and the percent Se-enriching rate was 58.3%. According to the analysis, the microbe was identified as a strain of Bacillus subtilis.
