摘 要:目的探讨MAPK、CyclinD1蛋白及其mRNA在病理性瘢痕及瘢痕癌组织中的表达及意义。方法①采用免疫组织化学(SP)法和原位杂交技术分别检测正常皮肤表皮,病理性瘢痕被覆上皮和瘢痕癌癌组织中MAPK及其mRNA和CyclinD1及其mRNA的表达。②利用计算机图像分析系统采集、分析图像,并对数据进行统计学处理。结果①MAPK及其mRNA和CyclinD1及其mRNA在瘢痕癌组织中呈强阳性表达,在病理性瘢痕上皮中呈弱阳性表达,在正常皮肤表皮呈阴性或弱阳性表达。瘢痕癌组表达水平、表达强度与正常皮肤表皮、病理性瘢痕被覆上皮组比较,差异均有统计学意义(P<0.01);但正常皮肤表皮组与病理性瘢痕被覆上皮组比较,差异无统计学意义(P>0.05)。②在瘢痕癌组织中,MAPK及其mRNA、CyclinD1及其mRNA、CyclinD1与MAPK蛋白、CyclinD1mRNA与MAPK mRNA的表达均呈正相关。结论①瘢痕癌中MAPK和CyclinD1的高表达与瘢痕癌的发生有相关性。②病理性瘢痕中MAPK、CyclinD1的表达与瘢痕上皮癌变没有相关性。Objective To study expression and significance of MAPK、MAPK mRNA and CyclinD1、and CyclinD1 mRNA in tissues of skin pathological scar and scar carcinoma.Methods ①The expression of MAPK 、MAPKmRNA and CyclinD1、CyclinD1mRNA were detected in normal skin epidermis,pathological scar epithelium and scar cancer tissues by the immunohistochemical method and in situ hybridization.② Pictures were collected and analyzed by the computer medical image analysis system,and SPSS statistic method was used to analyze the data.Results ① Expression of MAPK and CyclinD1 at both protein and mRNA levels showed negative or weakly positive in normal skin epidermis and pathological scar epithelium,but strongly positive in scar carcinoma tissues.The expression levels and intensity differences were statistically significant(P 0.01) as compared with normal skin group and skin scar group.There was no difference in MAPK and Cyclin D1 expression between normal skin group and pathological skin scar group(P 0.05).②Correlation analysis showed the goodcorrelations between the expression of MAPK and Cyclin D1,their mRNA expression and protein levels.Conclusion ①The high expression of MAPK and CyclinD in the scar cancer tissue is related to the occurrence of skin scar carcinoma.②The expression of MAPK and CyclinD1 in the pathological scar does not appear to be associated with scar carcinogenesis.
