胃癌组织中NF-κB、P73表达与幽门螺旋杆菌感染的相关性研究

摘　　要：目的:幽门螺旋杆菌(Hp)感染与胃癌发生发展有着密切而紧要相关,目前幽门螺旋杆菌被认为感染是诱发胃癌发生的重要因素之一。本组实验从胃癌组织中Hp与NF-κB、P73表达两者存在正相关性。方法:(1)收集2012-01～2013-03间佳木斯大学附属第一医院普外科胃癌手术住院病人100例。(2)Warthin-Starry银染法观察Hp感染情况,诊断是否存在幽门螺杆菌感染。(3)用免疫组化的方法检测样本中有无NF-κBP65及P73蛋白的表达。(4)采用SPSS10.0软件包进行统计分析。采用卡方检验,Fisher精确概率法及Spearman等级相关对相应资料进行分析,取α=0.05为检验水准。结果:有HP感染的胃癌组织、无HP感染的胃癌组织及癌旁组织的NF-κBP65蛋白阳性表达率分别为31.81%、20.58%和4.00%。差异有统计学意义(χ2=13.77,P<0.05);有HP感染的胃癌组织、无HP感染的胃癌组织及癌旁组织的P73蛋白阳性表达率分别为34.85%、17.65%和0。差异有统计学意义(χ2=22.23,P<0.05),正常的胃黏膜无NF-κBP65和P73蛋白的表达。结论:(1)胃癌患者NF-κB表达与幽门螺旋杆菌感染存在相关性。(2)胃癌患者P73蛋白表达与幽门螺旋杆菌感染存在相关性。(3)胃癌组织及癌旁组织均有NF-κB和P73的表达,而正常的胃黏膜组织没有NF-κB及P73的表达。Objective： Helicobacter pylori（Hp） infection and gastric cancer are closely related with each other.HP infection is now considered as one of the most important factors that trigger gastric cancer.In this set of experiments,there is a positive correlation between Hp in gastric cancer tissue and the expressions of NF-κ B and P73.Methods：（1）100 cases of gastric surgery inpatients were collected from January 2012 to March 2013,in General Surgery of the First Affiliated Hospital of Jiamusi University.（2） We observed Hp infection by Warthin-Starry silver staining,and diagnosed whether Helicobacter pylori infection exists.（3） We used immunohistochemistry to detect whether the expressions of NF-κBP65 and P73 protein exist in samples.（4） SPSS10.0 software package was used for statistical analysis.By means of the chi-square test,Fisher exact test and Spearman rank correlation,we analyzed corresponding data,taking α=0.05 for the test level.Results： The positive expression rate of NF-κBP65 protein in gastric carcinoma inflected by HP,in gastric carcinoma without HP inflection and adjacent tissues were 31.81%,20.58% and 4.00% respectively.In accordance with statistical comparison,the difference was statistically significant（χ2=13.77,P〈0.05）;the positive expression rate of P73 protein in gastric carcinoma inflected by HP,in gastric carcinoma without HP inflection and adjacent tissues were 34.85%,17.65% and 0.According to statistical comparison,the difference was statistically significant（χ2=22.23,P〈0.05）.There is no expression of NF-κBP65 or P73 protein in normal gastric mucosa.Conclusions：（1）There is a correlation between NF-κB expression of gastric cancer patients and Helicobacter pylori infection.（2） There is a correlation between P73 protein expression of patients with gastric cancer and H.pylori infection.（3） There are expressions of NF-κB and P73 in both gastric cancer tissue and cancer tissue,but no expression of NF-κB and P73 was existed in normal gastric mu
