摘  要:目的:检测实验性慢性胰腺炎(chronic pancreatitis,CP)大鼠造模后不同时间段内,骨髓间充质干细胞(mesenchymal stem cells,MSCs)的数量和增殖能力。方法:采用精氨酸腹腔注射法建立CP大鼠模型,并分别于造模后3、7、14 d获取大鼠骨髓细胞行原代培养,CFU-F计算克隆形成率。培养的细胞分别标记荧光抗体anti-CD29-PE/CY5、anti-CD44-FITC、anti-CD45-PE后,采用流式细胞仪检测MSCs细胞群CD29+CD44+CD45-占总体细胞的比例。结果:CP造模后3、7、14 d的原代培养骨髓细胞的克隆形成分别为(19.33±3.56)个、(24.67±4.89)个和(18.33±2.73)个,均显著高于对照组(4.83±1.72)个(P<0.01),获取的MSCs细胞CD29+CD44+CD45-百分比分别为(7.27±0.82)%、(7.94±2.11)%、(5.26±0.66)%,CP大鼠建模后3、7、14 d,获取MSCs比例显著高于对照组(3.37±0.55)%(P<0.05)。结论:CP大鼠建模后不同时间段MSCs数量和增殖能力发生改变,经历了先增加后减少的过程。Objective: To analyze the percentage and proliferation capacity of MSCs at different time points after CP induction. Methods: Experimental CP was induced in rats by intraperitoneal injection of L-arginine. Animals were sacrificed at 3 d, 7 d and 14 d after model induction. Bone marrow cells were implanted for primary cuhure. Colony formation rate of bone marrow ceils was calculated by CFU-F method. Surface markers CD29,CD44,CD45 on primary culture bone marrow cells were detected by flow cytometry,aud percentages of CD29+, CD44+CD45- cells (MSCs) in different groups were calculated. Results: Colony formation rates in 3 d,7 d and 14 d groups were 19.33±3.56,24.67± 4.89 and 18.33±2.73,respectively,significantly higher than that of control (4.83±1.72,P〈0.01). Percentages of CD29+, CD44, CD45-Cells (MSCs) were (7.27±0.82)%, (7.94±2.11)% and (5.26±0.66)%,respectively, significantly higher than that of control group (3.37+0.55)% (P〈0.05). Conclusions: The percentage and proliferation capacity of bone marrow MSCs changes during the course of CP, with a first increasing then reducing process.
