摘 要:目的研究沙格雷酯对野百合碱模型大鼠肺动脉高压的改善作用及对肺动脉活化T细胞核因子3(NFAT3)表达的影响。方法实验分为对照组、野百合碱诱导的肺动脉高压模型组、沙格雷酯干预组,每组6只大鼠。建立野百合碱诱导的肺动脉高压大鼠模型,沙格雷酯干预组给予5-HT2A受体拮抗剂沙格雷酯干预(100 mg/kg)4周。分别测定肺动脉压力,计算右心指数,肺动脉组织HE染色评价肺动脉壁增厚程度,Western blot检测肺动脉组织NFAT3蛋白表达水平。结果与肺动脉高压模型组相比,沙格雷酯干预组的平均肺动脉压力和右心指数降低,肺动脉壁增厚程度减轻,肺动脉NFAT3蛋白表达水平下调(P<0.05)。结论沙格雷酯可改善野百合碱动物模型的肺动脉高压程度,并下调肺动脉高压肺动脉的NFAT3蛋白表达水平。Objective To explore the effect of sarpogrelate on monocrotaline-induced experimental pulmonary artery hypertension and NFAT3 expression in pulmonary artery. Methods The rats were divided into control group( n = 6), model group ( n = 6) and sarpogrelate group(n =6). Pulmonary artery hypertension rat model was induced by intraperitoneal injection of monocrotaline (MCT). The model rats were daily given oral sarpogrelate (100mg/kg) for 4 weeks in sarpogrelate group. Pulmonary artery pressure was measured and right heart ratio was calculated. Pulmonary artery remodeling was evaluated by HE staining. NFAT3 protein level was measured by Western blot. Results Compared with model group, pulmonary artery pressure, right heart ratio and pulmonary artery remodeling de- creased in sarpogrelate group ( P 〈 0.05 ), and NFAT3 protein expression in pulmonary artery was also decreased ( P 〈 0.05 ). Con- clusion Sarpogrelate could improve the pulmonary artery hypertension in MCT-induced model and decrease the upregulated NFAT3 protein expression in pulmonary artery.
