摘  要:为探讨急性酒精性肝线粒体损伤和肝丙二醛(MDA)及超氧化物岐化酶(SOD)的变化,以小白鼠为材料进行了酒精灌胃实验。将20只小鼠随机分为对照组10只和实验组10只,实验组按体重10g/kg475mL/L酒精一次性灌胃,对照组灌胃生理盐水,灌胃后48h取肝作线粒体分离镜检和MDA、SOD检测。结果显示实验组肝线粒体长和宽分别为4.63μm和0.92μm,而对照组为3.31μm和0.72μm,P<0.01;实验组肝线粒体可见明显的肿胀和断裂;实验组肝MDA是1.63μmol/g,而对照组是0.74μmol/g,P<0.01;实验组肝SOD是3.70U/g,对照组是8.37U/g,P<0.01。实验结果表明,酒精可导致小鼠肝线粒体明显的损伤,肝MDA增高,SOD降低。In order to investigate mitochondrial damage and changes of malondidehyde and superoxide dismutase in acute alcoholic liver, 20 mice were randomly divided into the experimental group of 10 mice and the control group of 10 mice. The experimental group were given the alcohol(475mL/L) at the dose of 10g/kg body weight by gavages, and the control group were given the physiological saline. Animals were sacrificed after 48 h, and liver tissues were obtained. Malondialdehyde and superoxide dismutase levels were measured by spectrophotometry, mitochondria were extracted in homogenized liver tissues, and at last were examined by light microscope. The results indicated that the length and width of the mitochondria in mice hepatocyte were respectively 4.63 μm and 0.92 μm in the experi- mental group while those in the control group were 3.31 μm and 0.72 μm, P〈0.01. The mitochon- dria in mice hepatocyte had obvious swelling and rupture in the experimental group. The malondial- dehyde in mice liver was 1.63μmol/g in the experimental group while that in the control group was 0. 74 μmol/g, P〈0.01. The superoxide dismutase in mice liver was 3.70 U/g in the experimental group while that in the control group was 8.37 U/g, P〈0.01. In conclusion, alcohol can cause significant damage of mitochondria, the increase of malondialdehyde, and decrease of superoxide dismutase in mice hepatocyte.
