摘　　要：目的研究多肽AP25在组织水平和整体动物水平的抗肿瘤作用,为临床试验提供依据。方法采用大鼠动脉环和鸡胚尿囊膜实验检测AP25对血管新生的抑制作用,采用动物体内实验评价AP25抗肿瘤活性。结果大鼠动脉环实验表明多肽AP25可以抑制大鼠动脉环微血管生成,并且在0.25 mg·L-1时抑制率达到62.19%;鸡胚尿囊膜实验表明多肽AP25对血管新生有明显的抑制作用,且具有剂量依赖关系,20 mg·L-1抑制率达到80.37%;动物体内实验表明多肽AP25在裸鼠移植瘤模型上可以明显地抑制HCT116细胞的生长。结论多肽AP25对新生血管具有明显的抑制作用,主要通过抑制新生血管达到抑制肿瘤的生长,进而发挥抗肿瘤的活性。Aim To evaluate the anti-tumor effect of peptide AP25 by experiments on histology and animal level aiming at provide the basis for clinical trials. Methods The anti-angiogenic effect of AP25 was as- sayed by rat aortic ring experiment, chick chorioallan- toic membrane assay and the assay of tumor growth in- hibitory in nude mice. Results AP25 significantly in- hibited angiogenesis in rat aortic ring experiment and at the concentration of 0.25 was 62. 19% ; AP25 also mg ·L^-1, the inhibitory rate showed a dose-dependent inhibitory of angiogenesis in chick chorioallantoic mem- brane assay and at a concentration of 20 mg ·L^-1, the inhibitory rate was 80.37% ; AP25 significantly inhibi- ted the growth of HCTll6 cells in nude mice （P 〈 0. 01 ）. Conclusions It clearly suggests that AP25 has a strong anti-tumor activity. Its effect may depend on the inhibitory effect of angiogenesis.
