摘  要:目的探讨姜黄素预处理对大鼠局灶性脑缺血/再灌注损伤的影响。方法 SD大鼠(♂)56只,随机分7组,为假手术组、模型组(0.154 mol·L-1生理盐水5μl·g-1)、阳性对照组(尼莫地平0.4μg·g-1)和药物治疗组(姜黄素20、40、60μg·g-1)及溶媒对照组(DMSO),每组8只。药物治疗组在缺血前30 min分别腹腔注射相同体积的药物。局灶性脑缺血/再灌注损伤动物模型采用改良Longa线栓法制作。通过神经行为学评分、TTC染色法测定脑梗死面积和用实时定量RT-PCR对大鼠脑组织中BDNF mRNA的水平进行分析、酶联免疫吸附法(ELISA法)测定BDNF及其受体TrkB表达的方法,观察姜黄素预处理对大鼠脑缺血/再灌注损伤的保护作用。结果和模型组相比,姜黄素组大鼠神经缺失症状有明显改善(P<0.05),脑梗死面积减小(P<0.05),脑组织中BDNF mRNA、BDNF及其受体TrkB的含量(P<0.05)明显提高。结论姜黄素可降低行为学评分和脑梗死面积,增加大鼠脑缺血/再灌注损伤时BDNF mRNA、BDNF和其受体TrkB的表达水平,对脑缺血/再灌注损伤有保护作用。Aim To explore the effect of Curcumin pretreatment on injury induced by focal cerebral ische- mia reperfusion in rats. Methods 56 SD male rats were randomly divided into MCAO/R (middle cerebral artery occlusion ) model group, curcumin therapy groups( curcumin 20,40,60 μg·g^-1 ), the positive medicine control group ( nimodipine 0.4 μg·g^-1 ) and DMSO group. Each group has 8 rats. The same volume of drugs was intraperitoneally injected to therapy groups 30 minutes before cerebral ischemia. The MCAO/R model rats were made by thread-occluded method. The neurological deficiency scores were collected. Triphe- nyltetraazolium chloride(TTC) staining was used to e- valuate the infarct size. BDNF mRNA expression was tested by SYBR Green I real time RT-PCR. ABC- ELISA assessment was performed to evaluate the expression of BDNF and TrkB. Results Compared with MCAO/R model group, curcumin groups could obvi- ously improve some nervous deficit symptoms in the cerebral isehemia(P 〈 0. 05 ) and could reduce infarct size (P 〈 0.05). Besides, the expression of BDNF mRNA, BDNF and TrkB in the cerebral cortex after rat MCAO/R injury increased significantly ( P 〈 0.05 ). Conclusion Curcumin pretreatment has a neuropro- tective effect on focal cerebral ischemia-reperfusion in rats by improving neurological deficiency, reducing in- farct size and increasing BDNF mRNA, BDNF and TrkB amounts in the cerebral cortex.
