摘 要:目的检测透骨草提取物对人卵巢癌SKOV-3细胞增殖的影响,探讨透骨草提取物影响SKOV-3细胞增殖的相关分子机制。方法采用倒置显微镜观察透骨草提取物对SKOV-3细胞增殖的影响,采用免疫组织化学方法检测SKOV-3细胞P53蛋白和c-Myc蛋白的表达。结果倒置显微镜观察可见,37.8μg/mL透骨草提取物处理的SKOV-3细胞皱缩变形,轮廓不规则、不清晰,细胞脱壁增加。37.8μg/mL透骨草提取物处理的SKOV-3细胞P53蛋白和c-Myc蛋白的表达明显低于对照组(P<0.05)。结论透骨草提取物可以显著抑制SKOV-3细胞的增殖,其机制可能与下调P53蛋白和c-Myc蛋白表达有关。Objective To observe the effects of Phryma leptostachya L.var.asiatica Hara extract on proliferation of human ovarian cancer SKOV-3 cells and to reveal the related molecular mechanism.Methods The proliferation of SKOV-3 cells was observed with the inverted microscope.The expressions of P53 protein and cMyc protein of SKOV-3 cells were detected with immunohistochemistry technique.Results SKOV-3 cells treated with 37.8μg/mL Phryma Leptostachya L.var.asiatica Hara extract showed that SKOV-3 cells shrinked and deformed,the profile of SKOV-3 cells became irregular and unclear,and the number of exfoliative cells increased under inverted microscope.Compared with the control group,the expressions of P53 protein and c-Myc protein of SKOV-3 cells decreased significantly(P 0.05) after SKOV-3 cells were treated with 37.8μg/mL Phryma leptostachya L.var.asiatica Hara extract Conclusion Phryma leptostachya L.var.asiatica Hara extract had obvious inhibition effect on the proliferation of SKOV-3 cells and the molecular mechanisms may be associated with downregulating expressions of P53 protein and c-Myc protein of SKOV-3 cell.
