摘 要:目的探讨经内毒素(lipopolysaccharide,LPS)刺激不同时间及不同浓度谷氨酰胺(glutamine,Gln)预处理后对单个核细胞(peripheral blood mononuclear cell,PBMC)及热休克蛋白70(HSP70)表达的影响。方法取健康志愿者新鲜外周血滤过的血盘,用0.9%氯化钠注射液冲洗过滤,用密度梯度离心法提取PBMC并以LPS刺激不同时间(0、2、4、12、24 h),在LPS刺激2 h后使用不同浓度的Gln(0、1、4、8、10、15 mmol/L)预处理2 h,同时设空白对照组,分别检测PBMC中HSP70的表达。结果 LPS刺激2、4、12、24 h后HSP70表达均显著低于空白对照组,差异有统计学意义(P<0.05),LPS刺激12 h后的表达量显著高于2 h,差异有统计学意义(P<0.05)。HSP70的表达在Gln低浓度(1、4、8 mmol/L)预处理细胞培养后,与未经Gln预处理者比较差异无统计学意义(P>0.05);经浓度为10、15 mmol/L的Gln预处理细胞培养后,HSP70的表达较未经Gln处理者显著增强,差异有统计学意义(P<0.05);Gln浓度为10、15 mmol/L的预处理细胞HSP70的表达显著高于4 mmol/L Gln预处理细胞,差异有统计学意义(P<0.05)。结论 Gln可上调LPS诱导的PBMC HSP70的表达。Objective To explore the influence on expression of heat shock protein 70 (HSP70) in peripheral blood mononuclear cell (PBMC) after lipopolysaccharide (LPS) stimulation at different times and glutamine (Gln) pretreatment with different concentrations.Methods Fresh peripheral blood filtered through the blood discs of healthy volunteers were collected,then the blood on discs were flushed and filtered with 0.9% of sodium chloride injection,and PBMC was extracted by using density gradient centrifugation method with LPS stimulation at different times (0,2,4,12,24 h) respectively; 2 h of preconditioning was performed with different concentrations of Gln (0,1,4,8,10,15 mmol/L) after LPS stimulation for 2 h,and control group was also selected at the same time; expressions of HSP70 in PBMC of the two groups were detected respectively.Results Expressions of HSP70 in therapy group were significantly lower than those in control group after LPS stimulation for 2,4,12 and 24 h (P <0.05).After LPS stimulation for 12 h,the expression of HSP70 was significantly higher than that for 2 h (P <0.05).The expressions of HSP70 showed no significant differences between Gln low concentrations (1,4,8 mmol/L) of pretreatment and without pretreatment group (P > 0.05) ; the expressions of HSP70 showed significant differences between Gln high concentrations (10,15 mmol/L) of pretreatment and without pretreatment group (P < 0.05); The expression of HSP70 showed significant difference between 10 and 15 mmol/L Gln pretreatment and 4 mmol/L pretreatment (P < 0.05).Conclusion Gln may upregulate the expression of HSP70 in PBMC induced by LPS.
