摘 要:目的动态评价偏侧帕金森模型中实验猴的运动和睡眠状态,以了解帕金森病过程睡眠障碍的状况。方法以广西南宁灵康赛诺科生物科技有限公司提供的正常食蟹猴随机分为两组(对照组和实验组,每组4只)。给予老年食蟹猴单侧颈内动脉注射1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)(3 mg/只)建立偏侧帕金森病模型。用非人灵长类动物帕金森病模型行为学评分量表进行动态行为学评分,评价其运动障碍的严重程度。并在建模期间及建模稳定后用行为-身体活动监视器(PAMs)记录帕金森病模型猴睡眠状态的改变。结果 (1)建模成功后1.5年内行为学评分持续稳定,无显著波动,表明均已达到稳定的帕金森病状态。(2)注射MPTP后到成模前这一段时间日间睡眠时间及睡眠频次较对照组有显著增加(P=0.000、0.002),表明日间睡眠过度可能是帕金森病最早出现的睡眠异常。结论非人灵长类动物患帕金森病后出现夜间及日间睡眠时间延长且日间睡眠频次增多,表明在帕金森病过程中伴随有睡眠障碍的发生。Objective To evaluate the motor function and sleep disorders of monkeys as the Parkinson's disease(PD)model, in order to understand the status of sleep disorders in PD pathogenesis process. Methods Normal Macaca fascicularis which provided by the Guangxi Nanning the spirit Kangsainuoke Biological Technology Co., Ltd. were randomly divided into two groups (control and experimental group). In the experimental group, Old Macaca fascicularis were injected of MPTP( 1-methyl-4- phenyl-5-tetrahydropyridine, MPTP,3 mg each)into unilateral internal carotid artery to establish partial side of Parkinson's disease(PD) model and the "Explanation of Parkinsonian the Monkey Rating Scale" was used to evaluate their dynamic clinical scores and dyskinesia severity. Then PAMs was used to record the change of PD model monkey sleep states. Results ( 1 ) To 1.5 years after successful modeling the clinical score was steady, no significant fluctuations that have reached a stable state of PD. (2)Two years later, its motor function of clinical score declined than the success of model initial. In this period of time, before the PD model succeed after injected with MPTP, daytime sleep and sleep frequency compared with the control group, there was a significant increase( P = 0. 000,0. 002) which indicated that excessive daytime sleepiness may be abnormal in PD earliest sleep. Conclusions ( 1 ) The motor function barrier in modeling success 2 years later have improved than the initial stage of successful modal, maybe related to the compensatory function of dopaminergic neurons. (2) Increased daytime sleep time and its frequency are the earliest and the longest duration, appeared before the initial model which shows excessive daytime sleepiness occur at the preclinical and as early indicator of PD.
