摘  要:用免疫沉淀和免疫印迹法测定肿瘤相关抗原N - 35在不同肿瘤细胞及正常人心脏和肺组织的存在和分布情况 ;用N -聚糖酶酶解方法确定与单克隆抗体N - 35相对应的相关蛋白为一糖蛋白分子 ,不存在于正常人心脏及肺组织蛋白组分中 ,而以不同分子量形式分布于不同肿瘤细胞的蛋白组分中 .采用差速离心技术分离出肺腺癌细胞系GLC - 82亚细胞结构中的胞膜、胞核及线粒体成分 ,确认N - 35相关蛋白主要分布于胞核 ,线粒体次之 ,胞膜上分布量少 ,主要不以膜蛋白或跨膜蛋白的形式存在 ;经免疫荧光检测发现肿瘤相关抗原N - 35定位于肿瘤细胞有丝分裂S期的中心粒 (centriole)结构上 ,其功能很可能与肿瘤细胞无限增殖活动有关 .The relationship between the monoclonal antibody N-35 and the associated antigen expressed by Yunnan Gejiu pneumo-adenocarcinoma cell line GLC-82 and other caner cells wereidentified by the immunoprecipitation, immunoblotting, differential centrifigation and subcellary assay, mitotic cell immunoflourescence and N-glycanase digestion. The result showed that the protein molecule of the associated antigen N-35 was distributed among the Yunnan Gejiu human pneumoadenocarcinoma cell line GLC-82, human cervical cancer cell 1ine Hela, human hepatic cancer cell line HepG-2 and human breast cancer cell line PMC with different molecular weight, but no expression of the protein ingredient in normal human heart and lung fresh tissue. The molecular weigh of the associated antigen N-35 was 75~l30?KD. The associated antigen N-35 was determined as a glycanase digestion. with3 fragments of 65?KD,84?KD,86?KD. The differential centrifigation and subcellary assay showed the associated antigen N-35 distributed on the nucleus significantly much more than on the mitochondrail and membrane. The result showed by mitotic cell immunlflourescence that the associated antigen N-35 was located at the protein, and might be a very important tumor associated antigen N-35 was located at the centriole at the S Phage, suggesting strongly N-35 is mainly a nucleus controlling the malignant proliferation of cancer cells.
