摘  要:目的:采用双向电泳-质谱技术寻找TA2小鼠自发性乳腺癌发生相关的差异蛋白。方法:收集5只TA2自发乳腺癌小鼠和5只正常TA2小鼠血清,将血清标本进行双向电泳,经软件分析结合人工比对确认差异蛋白点,基质辅助激光解吸/电离飞行时间质谱对差异蛋白点进行鉴定。结果:在TA2自发乳腺癌小鼠和正常TA2小鼠血清间共找到11个差异蛋白质点,成功鉴定出5种蛋白质,它们分别为:妊娠区带蛋白、对氧磷酶、HemK家族修饰性甲基化酶、结合珠蛋白和血清淀粉样P成分。其中这5种蛋白均在TA2自发乳腺癌小鼠高表达,而在正常TA2小鼠呈低表达或表达缺失。结论:通过双向电泳质谱技术鉴定出的5种差异蛋白与乳腺癌的发生有一定关系,初步阐述了TA2小鼠自发乳腺癌发生的机制。Objective: To screen the differential protein related to spontaneous breast cancer in TA2 mice. Methods: Two-dimensional electrophoresis(2-DE) and matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS) were employed to screen the differential protein in the serum in 5 TA2 spontaneous breast cancer mice and 5 normal TA2 mice. Results: 2-DE serum maps for both groups were clear with high resolution.Eleven differential protein spots were founded and five proteins were successfully identified, such as pregnancy zone protein, paraoxonase, modification methylase HemK, haptoglobin, serum amyloid P componmnet. All of these pro- teins were highly expressed in TA2 spontaneous breast cancer mice, but much less or unexpressed in normal TA2 mice. Conclusion: Five serum differential proteins detected by 2-DE and MALDI-TOF-MS may lead to breast cancer and this paper also shows the mechanism of carcinogenesis in TA2 spontaneous breast cancer.
