摘 要:目的:建立大鼠肝细胞癌(hepatocellular carcinoma,HCC)细胞系C5F肝与脾原位接种模型,并比较其在不同内环境情况下对肿瘤发生和侵袭转移等能力的影响,筛选最佳C5F大鼠HCC肺转移动物模型。方法:20只雌性BALB/cA裸鼠随机分为肝、脾原位接种2组,观察裸鼠肝与脾成瘤率、裸鼠存活率、肿瘤病理学特点及远处脏器转移率及强度。结果:肝原位接种组成瘤率90%,肿瘤呈结节状膨胀性生长,肝与周围腹壁组织有粘连与侵袭,肺转移率80%。脾原位接种组脾成瘤率为56%,肝转移率67%,肺转移率33%。肝、脾接种组相比较,其肺转移率差异有统计学意义(P<0.05)。结论:C5F肝原位接种模型更适合于HCC发生及肺转移机制及药物干预等研究。Objective: To establish liver cancer models in nude mice by intrahepatic or in trasplenic injection of rat C5F hepatocellular carcinoma (HCC) cells, and to study the carcinogenic and metastatic phenotypes in the two models. Methods:C5F cells were injected into liver or spleen lobe of female BALB/cA nude mice. In the 5th week the tumor formation rate, carcinogenic features, and metastasis were comparatively studied. Results: In the live 90% animals developed liver group cancer with 80% lung metastasis. The tumor exhibited an expansive growing pattern with surrounding invasion. In the spleen group, 56% animals developed tumor, with a liver metastatic rate of 67%and lung metastatic rate of 33%. There was significant difference in the rate of lung metastasis between the liver and spleen models (P 〈 0.05). Conclusion: The liver in situ implant,model is more appropriato for the study of carcinogenesis,metastasis and medicinal intervention of HCC.
