抗PDGFR-ɑ抗体对铁锈诱导的人视网膜色素上皮细胞增殖过程的作用

摘　　要：目的:观察抗血小板源性生长因子受体ɑ(PDGFR-ɑ)抗体对铁锈诱导的人视网膜色素上皮细胞(hRPE)增殖活性的影响。方法:采用细胞体外培养技术,用铁锈建立hRPE细胞增殖分化模型。设置空白组、铁锈组、抗PDGFR-ɑ抗体(1,10,50,100μg/mL)处理组,分别作用0,12,24,48h后采用MTT比色法测定hRPE细胞的抑制率。结果:加入抗PDGFR-ɑ抗体后hRPE细胞增殖活性在一定浓度范围内随着抗体浓度增加而下降,而50μg/mL抗PDGFR-ɑ抗体为抑制hRPE细胞增殖的最佳浓度,其抑制率为42.44%。结论:抗PDGFR-ɑ抗体可以抑制hRPE细胞的增殖活性。AIM. To study the effect of platelet derived growth factor a receptor （PDGFR-a） antibody on the rust- induced human retinal pigment epithelial cell （hRPE ） proliferation. METHODS, In vitro rust hRPE cells proliferation differentiation model was established. Blank control group, rust group, PDGFR-a antibody （1, 10, 50, and 100 g/mL） treatment group were established. The effect of anti-PDGFR-a antibody on the growth of RPE was determined by MTT colorimetric assay after 0, 12, 24, 48h,and the inhibition rates were calculated accordingly. RESULTS.. After adding the anti-PDGFR-a antibody, hRPE cells proliferation activity was decreased at certain concentration as the antibody concentration increased, and 501Jg/mL anti-PDGFR-a antibody was the optimum concentration to inhibit hRPE cells proliferation, the inhibitory rate of hRPE was 42.44%. CONCLUSION： The anti-PDGFR-a antibody shows strong inhibitory effect on the growth of cultured hRPE.
