摘  要:目的:观察脑微血管内皮细胞(CMECs)对神经干细胞(NSCs)增殖分化的影响。方法:实验组原代培养CMECs和NSCs,鼠胚成纤维细胞(MEF)与NSCs共培养作为对照组,使用transwell体系共培养两种细胞7 d,观察NSCs的生长状况。移除CMECs,将NSCs置于分化培养基中4 d,观察其分化比例。结果:共培养期间,实验组较对照组NSCs数量更多,直径更大(P<0.001)(n=12);分化4 d后,实验组NSCs的神经元分化比例高于对照组(P<0.001)(n=6)。结论:CMECs能够促进NSCs的存活和增殖,且有助于提高NSCs的神经元分化潜能。Objective: To observe the effects of cerebral microvascular endothelial cells (CMECs) on the pro- liferation and differentiation of NSCs. Methods: The NSCs and CMECs were primarily cultured, purified and i- dentified. The survival and proliferation of NSCs were studied at day 7 after co-culture in transwells. After re- moving CMECs, NSCs were maintained in differentiation culture for 4 days to observe the differentiation rates. Mouse embryo fibroblast (MEF) cells co-cultured with NSCs were used as controls. Results: NSCs co-cultured with CMECs have increased the size and number ofNSCs (P〈0.001) (n=12) compared to those co-cultured with MEF cells. On day 4 after differentiation, NSCs showed a higher neuronal differentiation rates than those in the control groups (P〈0.001) (n=6). Conclusion: CMECs could promote the survival and neuronal differentiation po- tential of NSCs.
